摘 要:目的探讨姜黄素对子宫颈癌Hela细胞凋亡及对短的上腭、肺及鼻咽上皮克隆1(short palate lung and nasal epithelium clone 1,SPLUNC1)表达的影响。方法取对数生长期Hela细胞,分别设置对照组(姜黄素0μmol/L)、实验A组(姜黄素4μmol/L)、实验B组(姜黄素8μmol/L)、实验C组(姜黄素16μmol/L)、实验D组(姜黄素32μmol/L)、实验E组(姜黄素64μmol/L)、实验F组(姜黄素128μmol/L),采用四甲基偶氮唑蓝(MTT)法测定姜黄素对子宫颈癌Hela细胞的增殖抑制作用,4',6-二脒基-2-苯基吲哚(DAPI)染色观察细胞凋亡形态,异硫氢酸荧光素标记的膜联蛋白V/碘化丙啶(Annexin V-FITC/PI)双染流式细胞术检测细胞凋亡,Western blot检测SPLUNC1蛋白的表达。结果与对照组比较,实验各组细胞增殖抑制率均明显增加(P均<0.05),且除实验F组外,细胞增殖抑制率随姜黄素剂量增加而增加,呈剂量依赖性,差异有统计学意义(P均<0.05);其半数抑制浓度(IC50)=43.32μmol/L。对照组细胞形态正常,实验C、D、E组出现了典型的细胞凋亡形态变化,细胞核碎裂、固缩。与对照组比较,实验C、D、E组凋亡率随姜黄素剂量增加而增加,差异有统计学意义(P均<0.05);与对照组比较,实验C、D、E组SPLUNC1蛋白表达随姜黄素剂量增加而下降。结论在一定浓度范围内,姜黄素可明显促进子宫颈癌Hela细胞凋亡,抑制Hela细胞的增殖,其作用可能与下调SPLUNC1的表达相关。Objective To study the effects of curcumin on cervical cancer Hela cell apoptosis and expression of short palate lung and nasal epithelium clone 1 ( SPLUNC1 ) protein. Methods Hela cells in logarithmic growth phase were designed as control group ( cureumin 0 μmoL/L ) , experiment group A ( eurcumin 4 μmol/L ) , experiment group B ( curcumin 8 μmol/L) , experiment group C (curcumin 16 μmol/L) , experiment group D (curcumin 32 μmol/L), experiment group E ( curcumin 64μmol/L) , experiment group F ( cureumin128 μmoL/L). The 4-methyl thiazolyl tetrazolium (MTT) method was used to detect the inhibition effect of curcumin on cervical cancer Hela cells proliferation. 4' ,6-diamidino -2-phenyl indole(DAPI) staining was used to observe the morphological changes of cell apoptosis. AnnexinV-FITC/PI double staining flow cytometry was used to detect cell apoptosis rate. Western blot method was used to detect the relative expression level of SPLUNC1 protein. Results Compared with control group, the cell proliferation inhibitive rates in all experimental groups significantly increased ( all P 〈 0. 05 ) , and the cell proliferation inhibitive rates increased with the increase of eurcumin doses in a dose-dependent manner(all P 〈 0.05) except experiment group F. The 50% inhibitory concentration ( IC50 ) was 43.32 μmol/L. The cellular morphology in control group was normal, while typical apoptotic morphological changes of nuclear fragmentation and condensation appeared in experiment group C, D and E. Compared with control group, cell apoptosis rates increased with the increase of curcumin doses ( all P 〈 0.05) in experiment group C, D and E, and the expression levels of SPLUNC1 protein decreased with the increase of curcumin doses ( all P 〈 0.05 ) in experiment group C, D and E.Conclusions Curcumin could significantly promote apoptosis of cervical cancer Hela cell,and inhibit proliferation of Hela cell, and its effect may be associated with down-regulation
