摘 要:从四川省某规模化猕猴养殖基地的病死猕猴的脏器及肠道内容物中分离到1株可疑菌,对其纯化培养、革兰染色和生化鉴定后进行了分子水平鉴定,包括ipa H基因和wzy基因测定、16S r RNA基因序列分析,并进行了致病性分析及药敏试验。结果显示,该分离菌符合宋内氏志贺菌生物学及分子学特性,对小鼠具有较强致病性。16S r RNA基因多序列比对,绘制遗传距离及遗传发育树,证明其与2株宋内氏志贺菌(CP010829.1和HE616528.1)遗传距离最小,均为0.001。药敏试验显示,该菌株对头孢类等4类药物敏感,对青霉素类等6类药物耐药。结果表明,该分离株为宋内氏志贺菌且具有较强的致病性和耐药性。A strain of bacterium was isolated from stool specimens of diarrheal rhesus macaques (Macaca mulatta)in a large-scale farm located in Sichuan,China. The procedures of microbiological tests,including culture and purification,Gram-stain,biochemical tests,PCR-based detection of ipaH and wzy gene,pathogenicity test,antimicrobial susceptibility testing and 16S rRNAgene sequence anal- ysis,were used for isolation and identification of pathogen. The results demonstrate ipaH and wzy are positive,and the biochemical and biological features of the isolate correspond to Shigella sonnei's. The isolate has strong pathogenicity for mice. Compared with others,the genetic distances of 16S rRNA gene between the isolate and two strains of Shigella sonnei (sequence ID: CP010829.1 and HE616528.1) are minimal and both 0. 001. Furthermore,the isolate is sensitive to four kinds of antimicrobial includ- ing cephems,and is resistant to six kinds of antimicrobial including penicillins. In conclusion,the isolate is identified to be Shigella sonnei and displays strong pathogenicity and antimicrobial drug resistance.
