摘  要:目的研究Caveolin-1在非酒精性脂肪性肝炎(NASH)发病中的作用。方法对C57BL/6小鼠予高脂肪高胆固醇饮食建立NASH的动物模型,利用实时荧光定量PCR和Westernblot法检测小鼠肝组织中Caveolin-1基因和蛋白表达的改变。结果实验组小鼠在高脂肪高胆固醇饮食16周后全部发生非酒精性脂肪性肝病(NAFLD),与对照组比较,NASH组小鼠肝脏Caveolin-1的mRNA和蛋白水平均显著增高。结论Caveolin-1在高脂肪高胆固醇饮食性NASH小鼠的肝组织中表达增加,可能与NASH的发病机制相关。Objective To explore the role of Caveolin-1 in the progression of non-alcoholic steato- hepatitis (NASH). Methods C57BL/6 mouse model with NASH was established through feeding high fat and high cholesterol diet. Variation of expressed Caveolin-1 gene and protein in liver tissues of mice was de- tected with real-time fluorescence quantitative polymerase chain reaction (PCR) and Western blotting assay. Results The mice in the experimental group fed with high fat high cholesterol diet all developed non- alcoholic fatty liver disease (NAFLD) after 16 weeks, compared to the control group, the level of Caveolin- 1 mRNA and protein in the liver of mice with NASH were significantly enhanced. Conclusions The en- hancement of Caveolin-1 in liver of mice fed with high fat and high cholesterol diet may correlate with the occurrence of NASH.
