摘 要:目的 从瘤细胞液和瘤块两种植瘤方式中建立兔肾VX2 移植癌模型 ,并分析其病理学基础和DSA影像特征 ,为介入实验研究提供大型动物肾肿瘤模型。方法 3 0只新西兰白兔随机分两组 ,每组 15只。第一组 ,B超引导下将VX2 瘤细胞 (约 5× 10 7个 /ml)经皮注入左肾下极 ;第二组 ,B超引导下将瘤组织块 (约含 10 6~ 10 8个瘤细胞 )经皮注入左肾下极。之后 ,对两组进行比较观察 :①不同方式植瘤的成活率 ;②肾内肿瘤体积变化和肿瘤生长率 ;③大体及镜下 (光镜和电镜 )瘤组织形态特征 ;④成熟模型的肾动脉DSA表现。结果 ①两组植瘤成活率分别为 4/15、11/15 ,第二组植瘤成活率最高 (Ρ <0 .0 5 ) ,其瘤体呈指数性生长 ;②病理学表明该瘤在肾组织中呈浸润式生长 ,其性状与移植于兔肝的VX2 鳞状细胞癌特征相似 ;③DSA影像示移植性肾肿瘤具有丰富的血供 ,呈实体瘤表现。结论 成功建立了兔肾VX2 移植性癌模型 ,瘤块种植方式成功率明显高于细胞液注入法 ,为实体瘤介入治疗的实验研究提供了容易复制的大型动物模型。Objective To establish a stable VX 2 carsinoma model transplanted in rabbit kidney from two different transplanted methods of tumour cell fluid and tumour block fluid,and analyze its pathologic basis and image features (DSA) of the implanted tumors,to gain a reliablely mature larger tumor animal model for experimental study on interventional therapy of kidney carcinoma.Methods 30 white rabbits of New Zealand were divided into 2 groups at random,15 in each group.On the first group,VX 2 carcinoma cells (about 5×10 7/ml)were injected into the rabbit kidney lead by ultrasonic.On the second group,a piece of VX 2 tumor tissue(contained about 10 6~10 8 carcinoma cells)were injected into the rabbit kidney lead by ultrasonic.And then a comparative observation was performed in 2 groups included:①the success rate of implanted tumors;②the volume change and the growth rate of implanted tumors;③the biological features of implanted tumors in histopathology and DSA features of implanted tumors.Results ①The success rate of transplanted tumors in 2 groups were 4/15 and 11/15 respectively.The success rate of the 2nd group was obviously higher than the 2nd groups(Ρ<0.05),and in this group,the tumoral increasment was index number.②Histopathology indicated that the growth of implanted tumors was infiltrated into the rabbit kidney tissue,similar as the biological features of squamous cell carcinoma being transplanted in the rabbit liver.③DSA of the implanted kidney tumors displayed a hypervascular tumers.Conclusions It is successful to establish a retrofit model of VX 2 carcinoma transplanted into the rabbit kidney with a piece of tumor tissue,of which success rate is obviously higher than the other.This results make it possible to gain a reliablely mature larger tumor animal model for experimental study on interventional therapy of kidney carcinoma.
