循环microRNA对非小细胞肺癌诊断和化疗效果评价的研究

摘　　要：目的探讨血清中miR-19b,miR-125b,miR-34a,miR-200b的表达与非小细胞肺癌发生发展的相关性,以及这4种microRNAs在化疗过程中表达水平的变化和对患者化疗的监测效果。方法采用实时荧光定量PCR(Real-time PCR)技术检测43例健康体检者和非小细胞肺癌患者血清中miR-19,miR-125b,miR-34a,miR-200b的表达水平,以及化疗过程中的表达变化,比较第二次、第六次化疗后microRNAs的表达差异。结果与健康对照组相比,非小细胞肺癌患者血清中miR-19b,miR-125b和miR-200b表达上调,与淋巴结转移相关;miR-34a表达下调,与淋巴结转移无关。在化疗期间,miR-19b、miR-125b的表达随着化疗的进行而降低,第六次化疗后与第二次化疗后相比表达下降(P<0.05)。结论血清miR-19b,miR-125b,miR-34a和miR-200b可作为NSCLC的辅助诊断指标;miR-19b,miR-125b可作为NSCLC患者化疗效果评价的潜在标志物。Objective To explore the correlation between serum expression of miR-19 b,miR-125 b,miR-34 a,miR-200 bwith the development of non-small cell lung cancer,the expression changes of these four microRNAs during chemotherapy and the function of chemotherapy monitoring.Methods Real-time PCR was used to detect the expression change of serum miR-19 b,miR-125 b,miR-34 aand miR-200 bof 43cases healthy volunteers and non-small cell lung cancer patients.And also detected the expression changes of these four microRNAs during chemotherapy.To observe the differential expression between the second and sixth chemotherapy.Results Compared with the healthy control group,serum miR-19 b,miR-125 b,miR-34 aand miR-200 bexpressed differentially in patients of non-small cell lung cancer（P〈0.05）.The expression of miR-19 b,miR-125 b,miR-200 bwas up-regulated and related with lymph node metastasis.miR-34 awas down-regulated but not related with lymph node metastasis.The expression of miR-19 b,miR-125 bwere decreased when the patients received chemotherapy,and the expression were lower after the sixth cycle of chemotherapy than the second（P〈0.05）.Conclusion The serum expression of miR-19 b,miR-125 b,miR-34 aand miR-200 bcould be applied diagnostic markers for NSCLC;miR-19 b,miR-125 bcould act as potential biomarkers of chemotherapy evaluation for NSCLC patients.
