摘  要:目的观察纳米金(AuNPs)对荷结肠癌小鼠电刀手术前后小鼠血清细胞间黏附分子-1(ICAM-1)、转化生长因子-β(TGF-β)的影响。方法实验分为不同浓度AuNPs处理组和生理盐水对照组,建立荷结肠癌小鼠模型后,各组均经尾静脉注射0.2 ml相应的溶液,给药频率为2日1次,连续2周。然后经眶静脉取血,3 d后电刀手术切取各组小鼠移植瘤,检测移植瘤重量和抑瘤率。手术后3周经眶静脉取血,采用酶联免疫吸附试验(ELISA)法检测电刀手术前后小鼠血清ICAM-1、TGF-β浓度变化。结果手术前3 d各处理组荷结肠癌小鼠血清ICAM-1的浓度如下:空白对照组:(7.41±0.40) ng/ml,低浓度(2.5 μg/kg)AuNPs处理组:(5.32±0.35) ng/ml,高浓度(5.0 μg/kg)AuNPs处理组:(3.84±0.42) ng/ml(F=125.302,P=0.000)。手术后3周ICAM-1浓度为:假手术组:(15.46±1.71) ng/ml,单纯手术组:(3.44±0.37) ng/ml,低浓度(2.5 μg/kg) AuNPs+手术组:(1.76±0.61) ng/ml,高浓度(5.0 μg/kg)AuNPs+手术组:(0.92±0.33) ng/ml(F=313.424,P=0.000)。AuNPs对结肠癌小鼠血清TGF-β的影响与上述结果类似。此外,不同浓度AuNPs处理组结肠癌移植瘤重量及体积较空白对照组差异均有统计学意义(F=106.125,P=0.000),且呈剂量依赖关系。结论电刀手术前后AuNPs均能够降低荷结肠癌小鼠血清ICAM-1、TGF-β浓度,减小结肠癌小鼠移植瘤重量,发挥抗肿瘤作用。Objective To observe the influence of gold nanoparticles on intercellular adhesion molecule-1 (ICAM-1) and transforming growth factor-β (TGF-β) of colon carcinoma bearing mice before and after the electric knife treatment.Methods The experiment was divided into different concentrations of gold nanoparticles-treated groups and the saline control group. After the establishment of colon carcinoma bearing mice model, 0.2 ml solution in each group was injected via the tail vein every other day for 14 days. Then the transplanted tumors were taken out to measure the weight. The concentrations of serum ICAM-1 and TGF-β (3 days before and 3 weeks after the surgery) also were determined by enzyme linked immunosorbent assay (ELISA). Results Three days before surgery, the ICAM-1 concentration in 3 groups (control, 2.5 and 5.0 μg/ml gold nanoparticles) were (7.41±0.40), (5.32±0.35) and (3.84±0.42) ng/ml respectively (F=125.302, P=0.000). Three weeks after the surgery, the ICAM-1 concentration in 4 groups (sham operation group, simple operation group, 2.5 μg/ml gold nanoparticles+ operation group and 5.0 μg/ml AuNPs+ operation group) were (15.46±1.71), (3.44±0.37), (1.76±0.61) and (0.92±0.33) ng/ml respectively (F=313.424, P=0.000). Effects of gold nanoparticles on TGF-β concentration were similar to the above results. In addition, the xenograft weight in gold nanoparticles-treated groups was significantly different from that in the control group in a dose-dependent manner (F=106.125, P=0.000).Conclusion Gold nanoparticles can reduce serum ICAM-1 and TGF-β concentrations before and after the electric knife treatment, also reduce colon carcinoma mouse xenograft weight in vivo.
