摘  要:目的观察血管生长抑制因子(AS)和血小板内皮黏附分子34(CD34)在肝细胞癌组织中的表达,探讨AS和CD34与肝细胞癌发生、发展及预后的相关性。方法采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测肝细胞癌组织中AS和微血管密度(MVD)的表达,并分析其与肝细胞癌临床病理特征之间的关系。结果(1)正常肝组织AS阳性表达率为16.7%(2/12),肝细胞癌组织中其阳性表达率为58.0%(29/50)(χ2=6.150, P=0.002)。结果表明,肝癌组织中AS呈过表达状态。(2)在12例正常肝组织组织、50例肝癌组织中,其相应的MVD分别为25.70±5.20和33.72±4.35,两者相比较差异有统计学意义(P=0.000)。(3)肝细胞癌组织中AS的表达与患者的年龄(P=0.080)、性别(P=0.100)无明显相关性;而与肝癌组织的分化程度(P=0.000)、分期(P=0.002)、肿瘤直径(P=0.006)、门静脉是否受侵犯(P=0.000)以及AFP的表达水平显著相关(P=0.000);MVD与肿瘤组织分化程度(P=0.000)、分期(P=0.001)、肿瘤直径(P=0.002)、门静脉是否受侵犯(P=0.000),AFP的表达水平(P=0.000)明显相关。结论 AS在肝细胞癌中存在高表达,并且其和肝癌的分化程度、分期、肿瘤直径、门静脉是否受侵犯以及AFP的表达水平显著相关,进一步可产生抑制肝癌血管生成效应。Objective To observe the expression of angiostatin (AS) and platelet endothelial adhesion molecules 34 (CD34) in hepatocellular carcinoma (HCC) in order to investigate the correlation between AS and CD34 and the occurrence, development and prognosis of HCC. Methods The expression of AS and microvessel density (MVD) in hepatocellular carcinoma was detected by immunohistochemistry SP method, and the relationship between the expression of AS and MVD with the clinicopathological characteristics of HCC tissues was analyzed.Results (1) The positive expression rate of AS in normal liver tissues was 16.7% (2/12), and that was 58% (29/50) in HCC tissues with the difference being significant (χ2=6.150, P=0.002). The results showed that AS was overexpressed in HCC tissues. (2) In 12 cases of normal liver tissues and 50 cases of HCC tissues, the corresponding MVD expression rate was 25.70±5.20 and 33.72±4.35 respectively wtith the difference being statistically significant (P=0.000). (3) The expression of AS in hepatocellular carcinoma was not correlated with age (P=0.080) and sex (P=0.100). The expression level of AS in hepatocellular carcinoma tissues was related to the differentiation degree (P=0.000), tumor pathological stage (P=0.002), tumor diameter (P=0.006), whether the portal vein was violated (P=0.000) and also the expression level of AFP (P=0.000); The expression level of MVD in hepatocellular carcinoma tissues was also related to the differentiation degree (P=0.000), tumor pathological stage (P=0.001), tumor diameter (P=0.002), whether the portal vein was violated (P=0.000) and also the expression level of AFP (P=0.000).Conclusion AS is up-regulated in HCC tissues, and significantly correlated with the differentiation of HCC, staging, tumor diameter, invasion of the portal vein, and the expression of AFP level. AS can inhibit the angiogenesis in HCC.
