摘 要:为研究瘦素(leptin,LP)基因成熟肽第3外显子定点突变前后的结构差异,从而推测其功能差异,本试验根据GenBank公布的牛leptin序列设计特异性引物,扩增leptin成熟肽区域,并对leptin成熟肽序列进行克隆测序。结合生物信息学手段对牛leptin基因E3-125-C>A定点突变前后,leptin编码产物的结构和功能差异进行分析。结果显示,leptin单碱基的突变可改变氨基酸的编码,导致其在一级结构、二级结构形成显著差异,而三级结构相对较稳定。结果表明,leptin基因E3-125-C>A位置的单碱基突变,导致结构变化,可能从而引起功能变化。In order to study the differences in the structure of the mature peptide of leptin gene before and after the third exon site directed mutation, and speculated its functional differences, the mature peptide of leptin based on GenBank was amplified using specific primers. And the mature peptide of leptin was cloned and sequenced. The physical and chemical properties, structure and function of encoding products of leptin were compared and analyzed by bioinformatics methods. Leptin single base mutations could change the encoding of amino acids, and result in a sig- nificant difference in the primary structure and secondary structure, and the tertiary structure was relatively stable. Single nucleotide mutations at the E3-125-C〉A of leptin gene, caused structural changes that might cause functional changes.
