摘 要:目的研究比较TCF3-PBX1阳性和阴性急性淋巴细胞白血病基因表达的差异,并寻找在特定类型白血病的发生、发展过程中起重要作用的基因,为临床诊疗提供新思路。方法在公共基因芯片数据库GEO中下载GSE11877数据集,利用R编程语言,应用贝叶斯检验和倍比法(fold change)筛选差异表达基因(校正P<0.05,并且log2(fold change)>2)。利用DAVID数据库进行生物学过程功能富集分析。结果共获得207个数据集,其中TCF3-PBX1阳性组23个,阴性组184个,最终汇总获取2 890个基因的表达水平,其中27个符合条件的差异基因被选出,这些基因的功能大致分为细胞骨架结构、细胞信号转导、转录调控、细胞黏附、细胞凋亡等。功能富集分析显示BANK1、GAB1、ZAP70等8个基因在TCF3-PBX1阳性白血病发生、发展过程中可能起到关键作用。结论利用生物信息学方法能有效分析基因芯片数据并获取生物内在信息,TCF3-PBX1阳性白血病的发生是由于多种基因表达改变所致,为确定其早期诊断标志与新治疗靶位提供了新的思路。Objective To study and compare the differences of TCF3-PBX1 positive and negative acute lymphoblastic leukemia gene expression and to seek the gene for playing an important role in the development and progression process of specific type leukemia in order to provide a new idea for clinical diagnosis and treatment.Methods Microarray data set GSE11877 was downloaded from the public GeneChips database GEO.Differential expression genes were screened by using the R programming language,Bayes test and fold change method[correcting P〈0.05,log2(fold change)〉2].The biological processes function enrichment analysis was conducted based on DAVID database.Results A total of 208 data sets were obtained,including 23 data sets of TCF3-PBX1 positive and 184 data sets of negative.Finally 2 890 gene expression levels were obtained.In which 27 eligible differential genes were screened.These genes′functions were roughly divided into the cell skeletons construction,cell signal transduction,transcriptional control,cell adhesion and cell apoptosis.The function enrichment analysis displayed that 8genes(BANK1,GAB1,ZAP70,etc.)played the key role in the development and progression process of TCF3-PBX1 positive leukemia.Conclusion Using the bioinformatics method can effectively analyze the gene chip data and obtains the biological intrinsic information,the occurrence of TCF3-PBX1 positive acute lymphoblastic leukemia involves abnormal expression of multiple genes,which provides a new idea for determining early diagnostic mark and new therapeutic target.
