摘 要:目的:比较胆总管结扎(BDL)与CCl_4诱导小鼠肝纤维化的生化指标、肝脏病理学指标及α-Sma的表达。方法:60只健康雄性Bal B/C小鼠,分为CCl_4肝纤维化模型组(20只)及其对照组(10只)和BDL肝纤维化模型组(20只)及其对照组(10只)。观察小鼠一般情况,生化方法测血清ALT、TBA水平。HE和Masson染色观察肝组织的病理变化及免疫组织化学染色观察肝组织α-Sma的表达。结果:血清生化指标显示两组模型血清的ALT酶活性都明显高于对照组,肝组织的病理指标Masson染色及免疫组化检测均显示模型组的蓝色染色坏死区域明显多于对照组。结论:两种处理方法均可诱导小鼠肝纤维化,且BDL较早引起肝功能及肝纤维化指标升高。Objective To compare serum biochemical parameters,liver pathology,and α-Sma expression in Bal B/C rats with hepatis fibrosis induced by bile duct ligation( BDL) and carbon tetrachloride( CCl_4). Method Sixty healthy male Bal B/C rats were devided into CCl_4model( n = 20),CCl_4control( n = 10),BDL model( n = 20),BDL control( n = 10),General conditions of rats were examined,ALT,TBA were measured by biochemical analysis. Pathological changes in liver tissue were examined through hematoxylin eosin and Masson staining. Expression of α-Sma was assayed by immunohistochemistry. Results Serum biochemical analysis showed that ALT and TBA levels in two model groups were obvious higher than those control groups,Pathological changes in liver tissue( Masson staining and immunohistochemistry of α-Sma) also showed that the blue staining of two model groups were significantly higher than those control groups. Conclusion Both BDL and CCl_4 can induce hepatic in rats,The former induces early increase in the indicators of liver function and hepatic fibrosis.
