摘 要:[目的]检测食管鳞状细胞癌线粒体融合素基因-2(mitofusion2,Mfn2)基因启动子区域甲基化的情况,探讨其相关的临床意义。[方法]选取2014年2月~2016年3月我院收治的食管鳞状细胞癌患者116例组织蜡块,另取同期我院因食管良性病变切除的食管组织40例为对照组。Mfn2基因启动子区域甲基化使用亚硫酸盐修饰后PCR检测。[结果]食管鳞癌患者中Mfn2基因启动子区域甲基化率为37.1%(43/116),显著高于对照组的16.4%(19/116)(P〈0.05)。食管鳞癌患者男女性别和不同年龄之间Mfn2基因启动子区域甲基化率差异无统计学意义(P〉0.05)。T3、T4临床分期患者Mfn2基因启动子区域甲基化率为46.2%(24/52),显著高于T1、T2临床分期患者的29.7%(19/64)(P〈0.05)。有淋巴结转移患者Mfn2基因启动子区域甲基化率为44.4%(28/63),显著高于无淋巴结转移患者的28.3%(15/53)(P〈0.05)。肿瘤中、低分化患者Mfn2基因启动子区域甲基化率44.3%(31/70),显著高于高分化患者的26.1%(12/46)(P〈0.05)。[结论]Mfn2基因启动子区域在食管鳞癌组织中高甲基化,Mfn2基因启动子区域高甲基化与食管鳞癌的临床分期、肿瘤分化程度以及有无淋巴结转移密切相关。[Objective]To detect promoter region methylation of Mitofusion2(Mfn2)gene in esophageal squamous cell carcinoma and explore its clinical significance.[Methods]116cases of esophageal squamous cell carcinoma tissues were enrolled in this study.Another 40 cases of benign esophageal tissues were used as control.Mfn2 gene promoter was modified by bisulfite and methylation was detected using PCR analysis.[Results]The positive rate of Mfn2 gene promoter methylation in esophageal squamous cell carcinoma tissues was 37.1%(43/116),which was significantly higher than that of 16.4%(19/116)in control group(P〈0.05).The positive rate of Mfn2 gene promoter methylation was no significant difference between different gender and age in bladder cancer(P〈0.05).The positive rate of Mfn2 gene promoter methylation in T3 and T4stage group was 46.2%(24/52),which was higher than that of 29.7%(19/64)in T1 and T2stage group(P〈0.05).The positive rate of Mfn2 gene promoter methylation in lymph node metastasis group was 44.4%(28/63),which was higher than that of 28.3%(15/53)in non-lymph node metastasis group(P〈0.05).The positive rate of Mfn2 gene promoter methylation in median and low differentiation group was 44.3%(31/70),which was higher than that of 26.1%(12/46)in well differentiation group group(P〈0.05).[Conclusion]Hypermethylation of Mfn2 gene was detected in esophageal squamous cell carcinoma.Which was related to clinical stages,lymph node metastasis,and tumor differentiation of esophageal squamous cell carcinoma.
