摘 要:目的:研究灯盏花素(Bre)对动脉粥样硬化大鼠氧化应激和炎症反应的影响。方法:选取96只大鼠随机分为正常对照组,模型组,Bre 6、12、24 mg/(kg·d)组和辛伐他汀(SV)组(n=16),后5组采用高脂饲料喂养结合腹腔注射维生素D3的方法建立动脉粥样硬化大鼠模型,各治疗组造模过程同步灌胃给药,正常对照组给予正常饮食及腹腔注射等体积生理盐水;通过HE染色观察主动脉病理变化并进行病变分级;比色法测定血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及过氧化氢酶(CAT)活性和丙二醛(MDA)、活性氧簇(ROS)含量;酶联免疫法测定血浆中C反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-10(IL-10)含量。结果:与模型组比较,Bre干预组大鼠主动脉病理改变呈不同程度改善,Bre24 mg/(kg·d)组病变分级显著降低(P<0.01),Bre 12、24 mg/(kg·d)组血清中SOD、CAT活性显著升高(P<0.05),MDA、ROS含量显著降低(P<0.05),血浆中CRP、TNF-α、IL-1β及IL-6含量显著降低(P<0.05),Bre24 mg/(kg·d)组GSH-Px活性显著升高(P<0.05),IL-10含量显著升高(P<0.01)。结论 :Bre可能通过抑制氧化应激和炎症反应而改善动脉粥样硬化程度。Objective: To investigate the inhibitory effects of breviscapine (Bre) on oxidative stress and inflammation in atherosclerosis rats. Methods: Eighty rat models were made by dirtying with high fat and injecting VD3 and was divided into 5 groups: model group, Bre 6, 12, 24 mg/( kg· d) groups and 1.8 mg/( kg· d) SV group (n = 16), and the drugs were given by intragastric administration at the same time; setting normal control group (n = 16). The histopathological changes of abdominal aorta was observed by HE staining and graded; the activity of antioxidases and the content of MDA, and ROS in serum were determined; the levels of CRP, TNF-α, IL-1β, IL-6, and IL-10 in the plasma were determined with enzyme linked immunosorbent assay. Results: Compared with model group, the histopathological changes of the rats in Bre treated groups were improved, especially in Bre 24 mg/( kg · d) treated group (P 〈0.01); in Bre 12, 24 mg/(kg· d) treated groups, the activity of SOD, CAT in serum was significantly increased (P 〈 0.05 ), the contents of MDA, ROS in serum were significantly decreased (P 〈 0.05 ), and the levels of CRP, TNF-α, IL-1β, IL-6 in plasma were signifi-cantly decreased ( P 〈 0. 05 ) ; in Bre 24 rag/( kg· d) treated group, activity ot G^tt-Px in serum was significantly increased ( P 〈 0. 05 ) , and the level of IL-10 in plasma was significantly increased (P 〈 0. 01 ). Conclusion: Bre might mitigate atherosclerosis through restrain oxidative stress and inflamma- tion reaction.
