摘 要:目的从微生态与心血管损伤关系探讨清热解毒化浊片对心与动脉的影响。方法 70只SD大鼠随机分为正常组和造模组,造模组采用高脂、酒精复合因素干预12周建立肠源性内毒素血症(IETM)大鼠模型。造模成功后,按随机数列表法分为模型组、清热解毒化浊片组。经过4周治疗后采血检测内毒素(LPS)水平,观察心肌及主动脉组织病理变化,免疫组化法测定组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制剂(PAI)在心肌及主动脉组织中的表达。结果1)与正常组比较,模型组血浆LPS明显升高(P<0.01);心肌及主动脉组织PAI-1表达明显升高(P<0.01)、tPA明显减低(P<0.01);2)与模型组比较,清热解毒化浊片组,血浆LPS明显降低(P<0.01);心肌组织tPA表达升高(P<0.05),PAI-1表达明显降低(P<0.01);主动脉组织tPA表达明显升高(P<0.01),PAI-1表达降低(P<0.05);光镜下观察,清热解毒化浊片组大鼠心肌及主动脉炎性浸润情况明显改善。结论清热解毒化浊片可通过降低血浆LPS,协调tPA与PAI-1的平衡,保护肠道菌群失调导致的心与动脉损伤。Objective: To discuss the influence of Tablet Qingrejieduhuazhuo on heart and arteries from the re- lation between micro-ecology and cardiovascular damage. Methods: 70 SD rats were randomly divided into the normal group and the model group. Intestinal endotoxemia (IETM) rat models were established by intervention with high fat and alcohol composite factors for 12 weeks in the model group. The successful rat models were randomly divided into the model group and the Tablet Qingrefieduhuazhuo group. After 4 weeks of treatment,the endotoxin level was tested through blood sampling,the pathological changes in myocardium and aorta tissues were observed,the expression of tissue-type plasminogen activator (tPA) and plasminogen activator inhibitor (PAI) in myocardium and aorta tissues were measured by immunohistochemistry. Results: 1 )Compared with the normal group,plasma LPS increased significantly (P〈 0.01 ) in the model group ;expression of PAI-1 increased significantly (P〈 0.01 ),and that of tPA decreased significantly (P〈 0.01),in myocardium and aortic tissues ;compared with the model group,plasma LPS decreased significantly (P〈 0.01 ) in the Tablet Qingrejieduhuazhuo group ;ex- pression of tPA increased (P〈 0.05),and that of PAI-1 decreased significantly (P〈 0.01) in myocardial tissues; expression of tPA increased significantly(P〈 0.01 ), and that of PAI-1 decreased(P〈 0.05) in aortic tissues ; 2)The inflammatory invasion of myocardium and arteries in rats of the Tablet Qingrejieduhuazhuo improved significantly, observed under the light microscope. Conclusion: This study shows that Tablet Qingrejieduhuazhuo can protect myocardium and arteries damaged by flora disequilibrium in intestinal canal, by reducing plasma LPS and balancing tPA and PAI-1.
