摘 要:目的研究经多聚赖氨酸表面改性前后,兔同种异体骨对成骨细胞亲和力的变化,为提高同种异体骨的骨传导性能提供一个良好的方法。方法将取材于兔椎体的松质骨处理制成同种异体骨,采取冻干法,利用多聚赖氨酸对其进行表面改性。表面改性后的同种异体骨设定为实验组,表面未改性的设定为对照组,成骨细胞分别种植于2组同种异体骨的微孔表面。采用MTT法检测成骨细胞在同种异体骨微孔富集、增殖情况,同时检测2组成骨细胞表达的碱性磷酸酶(ALP)活性,并对2组结果进行比较。结果培养的第1—15天,2组细胞生长曲线呈抛物线形,细胞数量均持续增加,9 d后增殖速度减慢,11 d后进入数量增长平台期,且各时间点实验组成骨细胞数量均明显高于对照组(P均<0.05);在实验的4,8,12,16 d,实验组成骨细胞表达ALP活性均明显高于对照组(P均<0.05)。结论表面改性后的兔同种异体骨对成骨细胞的亲和力显著提升,利用多聚赖氨酸对同种异体骨进行表面改性是提高同种异体骨骨传导性能的一种理想方法。Objective It is to study the changes of affinity of rabbit allogeneic bone to osteoblasts before after surface modification with polylysine,thus to offer a good method to enhance the ability of osteoconduction of allogeneic bone. Methods The allogeneic bone was made from the cancellous of rabbits' centrum. The surface modification of the allogeneic bone was processed by the polylysine with lyophilization methods. The the allogeneic bone whose surface was modificated were taken as experimental group,and the one whose surface was not modificated were taken as control group. The osteobalsts were implanted onto the surface of the micropores in the two groups. The attachment and proliferation of osteoblasts on the surface of the micropores in the allogeneic bone were detected with MTT methods,and the activity of the ALP expressed by the osteoblasts were also evaluated respectively. Then the results were compared. Results In the culture stage from the first day to the fifth day,the curve of cell growth was parabolic,the numbers of cells continually increased,after 9 days the growth speed slowed down,and after 11 days reached stationary phase,the quantity of osteoblasts at every time point in experimental group was higher than that in control group( P〈0. 05). On the 4th,8th,12 th,16th day,the ALP activity in osteoblasts in experimental group was all significantly higher than that in control group( P〈0. 05). The attachment,proliferation of the osteoblasts in the experimental group were much superior than those in the control group( P〈0. 05). Conclusion The affinity of allogeneic bone to the osteoblasts can be significantly promoted after surface modification with polylysine. It is a good method that the surface modification of the allogeneic bone is processed by the polylysine to enhance the ability of osteoconduction.
