摘  要:利用细胞培养、蛋白激酶C(PKC)的抑制剂和免疫细胞化学法 ,探讨PKC对人低分化鼻咽癌细胞CNE 2Z中磷酸化酪氨酸蛋白分布的影响。结果发现 :磷酸化酪氨酸蛋白主要在胞膜 (6 5 2 % )与胞浆 (85 % )表达。作用于PKC调节区的抑制剂sphingosine(SS) (2× 10 -5mol/L)与作用于催化区的staurosporine(ST) (2× 10 -6mol/L)使其表达减弱、胞膜阳性率分别降低至 12 8%和 9 0 %。提示PKC对CNEProtein kinase C(PKC) plays a key role in modulating the growth of human poor differentiated nasopharyngeal carcinoma(NPC) cell line CNE 2Z.To further investigate its mechanism, the effect of PKC on the distribution of proteins phosphorylated on tyrosine in CNE 2Z cells treated with PKC inhibitors was determined with immunocytochemistry.The results showed that the proteins phosphorylated on tyrosine mainly located in cellular membrane and cytosol in CNE 2Z cells,and the positive cell percentage of expression in membrane decreased from 65.2% to 12.8% and 9.0% respectively after treated with sphingosine(SS)(2×10 -5 mol/L),a potent inhibitor acting on the modulating domain of PKC, and staurosporine(ST)(2×10 -6 mol/L),a potent inhibitor acting on the catalytic domain of PKC.The findings suggest that PKC may modulate the process of proteins phosphorylated on tyrosine in CNE 2Z cells.
