摘  要:目的:探讨micro RNA-221在早期结肠癌患者粪便中的表达情况,并研究其作为早期结直肠癌以及癌前病变分子诊断标志物的可行性。方法:采用实时荧光定量聚合酶链反应检测245例大肠病变患者的粪便及30例正常人群粪便中micro RNA-221的水平,分析micro RNA-221与不同临床病理分级的关系。结果:结肠病变的患者粪便中micro RNA-221水平随着病理分级增加而增高,差异具有统计学意义(P<0.001)。进展型腺瘤和早期结直肠癌组织中micro RNA-221水平与对照组相比明显升高,差异有统计学意义(P<0.001)。结论:早期结直肠癌粪便中micro RNA-221的表达上调,可能参与了结直肠癌的发生及发展,粪便micro RNA-221有可能成为筛查早期结直肠癌非侵入性的标志物。Objective The detection of microRNA (microRNANA) dysregulation in stool is a novel approach for the diagnosis of early colorectal cancer. The aim of this study is to investigate the use of microRNA-221 in stool samples as non-invasive biomarker for early colorectal cancer diagnosis. Methods The levels ofmicroRNA-221 in 245 stool samples with colorectal lesions were measured by real-time PCR. The relationship between the expression ofmicroRNA-221 and clinicopathological features was analyzed. Results In stool samples, the level of microRNA-221 showed a significant increasing trend with the increase of pathological grade(P 〈 0.001).The level of microRNA-221 in tissue of colorectal lesions was statistically higher than in the controls(3.02 ± 0.68 VS 1.10 ±0.19, P 〈 0.001 ). Conclusion The over-expressd microRNA-221 might play a role in the development and progression of colorectal carcinoma. Stool-based microRNA-221 can be used as a non-invasive biomarker for prognosis of early colorectal cancer.
