摘 要:目的本文以胆汁酸类成分为例,研究中药系列化学成分在甲醇-乙腈-水三元流动相系统中的色谱保留行为。方法采用LTQ-Orbitrap检测器,研究三元流动相系统中6种胆汁酸类成分的反相色谱保留时间与甲醇/乙腈(m)、乙腈/甲醇(n)的关系。结果在以乙腈为主的三元系统中,水相比例为40%,0.43<m<1时;或在以甲醇为主的三元系统中,水相比例为25%,0<n<0.11时,胆汁酸类成分可获得良好的基线分离。结论甲醇-乙腈-水三元系统可作为分离复杂体系中胆汁酸类化学成分的新型反相色谱流动相。Objective To investigate the RP-LC retention behavior of cholic acids in three-component mobile phase comprising methanol,acetonitrile and water.Methods LTQ-Orbitrap was used as the detector to systematically study the relations between the retention time of six cholic acids and the ratios of methanol/acetonitrile(m)or acetonitrile/methanol(n).Results Base-lined separation of cholic acids could be achieved in an acetonitrile-dominated three-component mobile phase when the proportion of water was40% and 0.43m1,or in a methand-dominated three-component mobile phase when the proportion of water was 25% and 0n0.11.Conclusion The three-component phase provides a new solution to RP-LC separation of cholic acids in complex systems.
