肿瘤患者的体外药物敏感试验

摘　　要：目的　探讨 3- (4,5 ) -双甲基 - 2噻唑 - (2 ,5 ) -二甲基溴化四氮唑蓝 (MTT)体外药敏法测定肿瘤细胞对化疗药物的敏感性。方法　 113例肿瘤患者进行了体外药敏试验。结果　肺癌患者对CBP、VM2 6、THP、DDP、BLM、MTX、HCPT较敏感 ,对VP16、DTIC、EADM、VCR不敏感 ;乳腺癌患者对CBP、BLM、DDP、5 -FU、HCPT、MTX、THP、MMC较敏感 ,对VM2 6、VCR、VP16、TAX不敏感 ;神经胶质瘤患者对VM2 6、DDP、5 -FU、MMC、ACR、ADM、BLM较敏感 ,对CCNU、MTX、VCR、PCB不敏感。不同个体对同一药物的敏感性以及不同阶段相同药物对不同个体的敏感性具有明显差异 ,不同类型的肿瘤有着不同的药敏谱。结论　化疗药物的敏感性检测对临床选择化疗药物具有重要的参考价值。MTT法为临床肿瘤化疗的药物敏感性检测提供了简便。OBJECTIVE To study the anti-cancer drug sensitivity in tumor patients with MTT method.METHODS Chemosensitivity testing for 12～15 drugs was performed on fresh tumor samples from 45 cases of lung cancer patients, 32 cases of breast cancer and 36 cases of neuroglioma by MTT method.RESULTS Lung cancer was sensitive to the chemotherapeutic drugs CBP, VM26, THP, DDP,BLM, MTX and HCPT, nonsensitive to VP16, DTIC, EADM, VCR;breast cancer was more sensitive to the chemotherapeutic drugs CBP, BLM, DDP, 5-FU, HCPT, MTX, THP, MMC,nonsensitive to VM26, VCR, VP16, TAX;neuroglioma was more sensitive to VM26, DDP, 5-Fu, MMC, ACR, ADM, BLM,nonsensitive to CCNU, MTX, VCR, PCB. The results showed that the chemosensitivity of an individual to a given drug differs significantly from another individual,and the chemosensitivity of different individuals to the same drug can be rather different at various therapeutic periods.Chemosensitivity spectra vary with tumor types.CONCLUSION The MTT method is very rapid and simple to perform.It can be applied to clinical drug sensitivity testing of human tumors.
