针刺对去势雌兔干眼症模型眼表粘蛋白MUC5AC、MUC19表达的影响

摘　　要：目的研究针刺对更年期干眼症模型去势雌兔眼表蛋白MUC5AC和MUC19的表达影响,为更年期干眼症治疗提供依据。方法选择健康成年雌兔45只,将其随机分为对照组、模型组、针刺组(每组15只),模型组和针刺组行双侧卵巢切除建立干眼模型,2周后对针刺组采取针刺治疗,分别于去势前、2周后、3周后、6周后行Schirmer试验、泪膜破裂时间检测、荧光染色评分、虎红染色评分、免疫组化染色检测MUC5AC的MUC19表达水平。结果 2周和3周时模型组和针刺组Shirmer值、BUT时间、荧光染色评分、虎红染色评分值与对照组相比,差异有统计学意义(P<0.05);3周时针刺组和模型组的Shirmer值和虎红染色评分差异有统计学意义(P<0.05);6周时模型组的Shirmer值、BUT时间、荧光染色评分、虎红染色评分值与对照组和针刺组相比,差异有统计学意义(P<0.05)。实验3周后针刺组MUC5AC和MUC19表达水平较模型组显著升高(P<0.05),与对照组相比差异有统计学意义(P<0.05);6周后针刺组黏蛋白表达水平继续升高,明显高于模型组(P<0.05)。结论针刺治疗可提高眼表黏蛋白的表达,提示其在干眼症治疗中具有应用价值。Objective To investigate the effect of acupuncture on the expression of MUC5 AC and MUC19 in castrated female rabbits model of dry eye symptom after menopause. Methods A total of 45 healthy adult female rabbits were randomly divided into control group,model group and acupuncture group（ 15 rabbits each group）. The ovariectomy surgery was performed in the model group and acupuncture group to produce dry eye models. The acupuncture treatment was performed in the acupuncture group within 2 weeks after the surgery. The Schirmer test,tear film break-up time（ BUT）,fluorescein staining,rose bengal staining and immunohistochemical staining were performed to test MUC5 AC and MUC19 expression before the surgery and 2（ w2）,3（ w3） and 6（ w6） weeks after the surgery. Results At w2 and w3 after the castration surgery,Shirmer value,BUT value,fluorescein staining scores and rose bangle staining scores of both model group and acupuncture group were significantly different from those of the control group（ P〈 0. 05）. At w3,there was a significance difference in Shirmer value and fluorescein staining scores between model group and acupuncture group（ P〈 0. 05）. At w6,Shirmer value,BUT value,fluorescein staining scores and rose bangle staining scores of model group were significantly different from those in the control group and puncture group（ P〈 0. 05）. On w3,the expression of MUC5 AC and MUC19 in the acupuncture group increased obviously as compared with model group（ P〈 0. 05）,and the levels of MUC5 AC and MUC19 in the two groups were different from those of the control group（ P〈 0. 05）. Six weeks later,the levels of mucins in the acupuncture group were higher than those of the model group（ P〈 0. 05）. Conclusion The acupuncture treatment can promote the expression of MUC5 AC and MUC19 mucin on ocular surface in rabbits with dry eye symptom after menopause,suggesting its role in the treatment of dry eye symptom.
