摘  要:目的通过观察内质网应激(ER Stress)通路相关分子葡萄糖调节蛋白78(GRP78)及CCAAT/增强子结合蛋白同源蛋白(CHOP)在新生小鼠巨细胞病毒肝炎中的表达变化,探讨ER Stress在巨细胞病毒肝炎发生、发展中的作用。方法将64只新生小鼠随机分为对照组(n=32)与病毒组(n=32)。病毒组于小鼠出生3天时腹腔接种MCMV病毒悬液20μl(107.21μ/0.1 ml),建立小鼠巨细胞病毒肝炎模型;对照组于相同时间点腹腔注射等量的无菌生理盐水,分别于3 d、7 d、14 d、28 d时处死小鼠。ELISA法检测小鼠血清中丙氨酸转氨酶(ALT)的水平,RT-PCR法检测小鼠肝细胞内GRP78、CHOP的mRNA水平,免疫组化法检测小鼠肝细胞内GRP78、CHOP的表达及分布情况。结果成功建立小鼠巨细胞病毒肝炎模型;各时间点病毒组GRP78、CHOP的mRNA及蛋白表达均较对照组显著增加,差异均有统计学意义(P<0.05)。结论 ER Stress相关分子GRP78和CHOP在新生小鼠巨细胞病毒肝炎中表达均明显增加,说明新生小鼠巨细胞病毒肝炎中发生了ER Stress,ER Stress可能在新生小鼠巨细胞病毒肝炎中发挥重要作用。Objective To observe the changes of expressions of glucoge-regulating protein 78(GRP78) and CCAAT/enhancer-binding protein homologous protein(CHOP) in neonatal mice with murine cytomegalovirus hepatitis,explore the role of endoplasmie retieulum stress(ER Stress) in occurrence and development of cytomegalovirus hepatitis.Methods Sixty-four neonatal BALB/c mice were randomly divided into MCMV group and control group randomly,32 mice in each group.The neonatal mice in MCMV group were treated with intraperitoneal inoculation of 20 μl MCMV suspension(TCID50:107.21/0.1 ml) on the third day after birth to establish MCMV disseminated infection model;the neonatal mice in control group were treated with transabdominal injection of sterile normal saline of the same dose.The mice after three,seven,fourteen and twenty-eight days were killed.ELISA was used to detect the levels of serum alanine aminotransferase(ALT);semi-quantitative RT-PCR was used to detect the expressions of hepatic GRP78 CHOP mRNA,immunohistochemical staining was used to detect GRP78 and CHOP levels in hepatic tissue.Results The model of neonatal mice with murine cytomegalovirus infection was successfully established,the expression levels of hepatic GRP78 CHOP in MCMV group were statistically significantly higher than those in control group(P〈0.01).Conclusion The expression levels of ER stress related molecules GRP78 and CHOP in neonatal mice with murine cytomegalovirus hepatitis increase.The mice with murine cytomegalovirus hepatitis undergo ER stress.ER stress might play an important role in murine cytomegalovirus hepatitis.
