摘 要:目的:探讨敲减及过表达14-3-3β基因对胃癌细胞生物学行为的影响及临床病理改变。方法在 SGC 7901、MGC 803和 HGC 27胃癌细胞系中,利用 RNA 干扰技术抑制14-3-3β基因的表达,分别用 MTT 法、基底膜侵袭实验检测胃癌细胞的增殖和侵袭能力的变化。结果在 HGC27和 MGC803细胞系中,14-3-3β基因的缺失会影响细胞增殖( P 值分别为0.000、0.000、0.000、0.000、0.000、0.001、0.000、0.005),14-3-3β基因表达缺失的MGC803胃癌细胞侵袭能力降低( P 值分别为0.000、0.000)。结论14-3-3β基因参与了胃癌细胞增殖和侵袭。Objective To investigate the biological behavior and the clinicopathological signifi -cance with silencing of 1 4-3-3βexpression in gastric cancer .Methods RNA interference method was used to knock-down the expression of 1 4-3-3βin gastric cell lines , and MTT method was used to detect tumor cell proliferation .We used transwell test to check the invasive ability of gastric tumor cells without 1 4-3-3βexpression .Results RNA interference could downregulate the endogenous 1 4-3-3βexpression efficiently in MGC 8 0 3 and HGC 2 7 cell lines .After transient transfection in HGC 2 7 and MGC 8 0 3 cell lines .The proliferation rate of transfected cells was significantly decrease compared to the negative control ( P values are 0.0 0 0 , 0.0 0 0 , 0.0 0 0 , 0.0 0 0 , 0.0 0 0 , 0.0 0 1 , 0.0 0 0 , 0.0 0 5 ) .Furthermore , the transwell chamber test showed that the absence of 1 4-3-3βexpression in MGC 8 0 3 could reduce the invasive capability of cancer cells ( P values are 0.0 0 0 , 0.0 0 0 ) . Conclusion RNA interference method is effective in koncking-down 1 4-3-3βexpression in gastric carcinoma cell line .1 4-3-3βmay be involved in regulating cell proliferation and invasion of gastric cancer .
