摘 要:目的观察弓形虫ROP16蛋白转染肝癌细胞Hep G2后,肝癌细胞的凋亡比率的变化以及细胞凋亡蛋白的表达水平变化。方法克隆弓形虫ROP16基因,构建真核转染质粒,并用脂质体转染的方式转染Hep G2细胞,48小时后,用流式细胞仪检测细胞凋亡比率,用western blotting和RT-PCR的方法检测细胞凋亡相关蛋白caspase-9、Bax、Bcl-2等的表达水平。结果与未转染细胞组相比,弓形虫ROP16转染Hep G2细胞后,肝癌细胞的凋亡比率明显升高。促细胞凋亡蛋白caspase-9、Bax的蛋白表达水平和基因转录水平均升高,而抑制细胞凋亡蛋白Bcl-2的表达水平和转录水平下降。结论 ROP16蛋白可以诱导肝癌细胞Hep G2发生凋亡。Objective To investigate whether ROP16 could induce apoptosis of liver cancer cell line HepG2 and expression profile of apoptosis related proteins. Methods 48 hrs after transfection of HepG2 with plasmid of pROP16, apoptotic cell rate was detected by FACS. Transcription level or expression level of caspase-9, Bax and Bcl-2 were evaluated via real-time PCR or western blot- ting. Results Compared with control group, apoptotic rate of transfected HepG2 was slightly high. And expression level of caspase-9 and Bax were significantly higher, but Bcl-2 level was lower. Conclusions ROP16 could induce apoptosis of HepG2 and change ex- pression levels of apoptosis related proteins.
