摘  要:将酸胁迫(p H=3)下的马铃薯脱毒试管苗转接于不适合试管薯诱导的MS培养基(含蔗糖30g/L)中,进行试管薯诱导,并对试管薯诱导过程中植株的蔗糖和淀粉含量及蔗糖合酶(sucrose synthase,SS)、可溶性淀粉合成酶(soluble starch synthase,SSS)和腺苷二磷酸葡萄糖焦磷酸化酶(ADP-glucose pyrophosphorylase,AGPase)的活性变化进行研究。结果表明,经酸胁迫的植株均可缩短试管薯诱导时间,经酸胁迫1d的植株在第13天开始形成试管薯,诱导时间最短,单薯重为187.8mg,单株结薯数量为2.2个,而不经酸胁迫的植株未能诱导出试管薯。植株在受酸胁迫期间,叶片中蔗糖含量无明显变化,而SS、SSS和AGPase的活性显著升高;在试管薯诱导过程中,酸胁迫植株叶片的SS活性呈降低和升高交替趋势;SSS活性在诱导试管薯0~5d时呈先下降后上升的趋势,之后趋于稳定;AGPase活性呈先上升后下降、之后再上升的趋势。The transfer of in-vitro potato plantlets in pH 3 solution to the MS medium containing 30g/L sucrose which was against formation potato tuber was conducted to induce the microtuber and evaluate the contents of sucrose and starch potato plantlets as well as the changes in sucrose synthetase (SS), soluble starch synthetase (SSS) and ADP- glucose pyrophosphorylase (AGPase) activity during the induction. Results indicated that a short induction time and the highest number of potato microtubers occurred from potato plantlets under stress, however untreated plantlets failed to induce tubers.For microtuber formation after pH 3 stress for ld, microtuberization starts at the 13th day in MS medium, and the number of microtubers for a single plantlet was 2.2 and the mean weight of microtuber was 187.8mg. No significant change was found in sucrose content, but SS, SSS, and AGPase activities increased significantly in leaves during acid stress treatment. SS activity decreased and increased alternately, SSS activity decreased and then increased at 0-5d, but later tended to stabilize. AGPase activity in leaves of induced microtuber tended to increase and then decrease but later increased in leaves ofplantlet with acid stress during inducing microtuber.
