摘 要:目的建立放射性粒子离体照射模型,观察^125I粒子持续低剂量率照射对人舌鳞癌CAL-27细胞的抑制效果。方法建立放射性粒子环形排布的照射模型,将指数生长期细胞分为4组:空白对照组、不同剂量粒子照射组(2、4、6Gy组),采用CCK-8法检测^125I粒子照射对人舌鳞癌CAL-27细胞的抑制率,倒置显微镜观察细胞学形态变化。结果2、4、6Cy粒子照射组所对应的细胞抑制率分别为(60.27±1.07)%、(71.48±0.65)%和(76.59±1.50)%,经方差分析不同照射剂量组间抑制率差异有统计学意义,呈时间和剂量依赖性(F=314.475,P〈0.01)。结论本方法建立的放射性粒子离体照射模型简便实用,可用于开展放射生物学实验,为研究^125I粒子持续低剂量率照射对舌鳞癌细胞的作用机制奠定了基础。^125I放射性粒子对CAL-27细胞生长具有显著的抑制作用。Objective To establish a model of ^125I seed irradiation in vitro and investigate its inhibitory effects on human tongue squamous carcinoma cell line CAL-27. Methods The radiation model of radiative particles in a circular array was established. The cells in the exponential phase were divided into four groups: a blank group and irradiation groups using various doses of radiative particles ( Groups 2, 4, and 6 Gy). Then, CCK - 8 assay was performed to evalu- ate the inhibitory effects of ^125I seed irradiation on CAL- 27 cells. Meanwhile, the morphological changes were observed under an inverted phase contrast microscope. Results The inhibitory rate of Groups 2, 4, and 6 Gy was (60.27±1.07 ) % , (71.48±0.65 ) % and (76.59 ± 1.50) % , respectively. According to analysis of variance, these inhibito- ry rates were statistically different in a time - and dose - manner (F = 314. 475, P 〈 0.01 ). Conclusions The radia- tion model of radiative particles is simple and efficient for radiobiological experiments, which lay foundations for studies a- bout the mechanism underlying the inhibitory effects of 125I seed irradiation on CAL -27 cells, ^125I seeds can remarkably inhibit the proliferation of CAL -27 ceils.
