摘  要:目的探究甲基化抑制剂5-氮杂-2'脱氧胞苷(5-Aza-Cd R)对肺癌A549细胞株DNA甲基转移酶1(DNMT1)及c-myc、MKi-67、p53基因表达的影响。方法选取合适的肺癌A549细胞株作为研究对象,随机将这些细胞分为观察组和对照组,采用不同的方法进行处理,其中观察组采用5-Aza-Cd R进行处理,在对照组的细胞株中不加任何的药物处理,经过3d的药物处理之后,主要采用FQ-PCR的检测技术研究5-Aza-Cd R对肺癌A549细胞株DNMT1及c-myc、MKi-67、p53基因表达的影响。结果研究发现观察组的DNMT1及c-myc、MKi-67m RNA与对照组相比,表达量明显的低于对照组,但p53 m RNA的表达量比对照组高,P<0.05有统计学意义。结论采用甲基化抑制剂5-Aza-Cd R明显的降低了肺癌A549细胞中的DNMT1及c-myc、MKi-67、p53基因表达量,使得p53基因表达量升高,主要的原因可能与基因启动子区域的去甲基化相关,为去甲基化的治疗提供了有利的线索及依据。Objective To explore the effect of methylation inhibitor 5-Aza-CdR on the expression of A549, c-myc, MKi-67 and p53 genes in lung cancer cell line DNMT1. Methods The A549 lung cancer cell suitable line was regarded as the research object, and these cells were randomly divided into observation group and control group. The observation group used 5-Aza-CdR treatment, and the cells in the control group didn't receive any drug treatment. After 3 days treatment, FQ-PCR detection technology was used to research the effect of 5-Aza-CdR on A549 lung cancer cell lines DNMT1 and c-myc, MKi-67, p53 gene expression. Results The study found that the expression of c-mye and MKi-67mRNA, DNMT1 of the observation group was significantly lower than the control group. But the expression of mRNA p53 was higher than the control group, and P 〈 0.05 had statistical significance. Conclusion The methylation inhibitor 5-Aza-CdR significantly decreases lung cancer A549 cells in DNMT1 and c-myc, MKi-67, p53 gene expression, and the gene expression of p53 increases. The main reason may be related with sub region demethylation related gene promoter. This provides clues and basis for favorable treatment for demethylation.
