摘 要:目的:探讨淫羊藿醇提物(EEBM)对高脂血症模型小鼠血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)水平的影响,阐明其抗氧化作用。方法:采用铁离子还原力法、2,2-二苯基苦基肼(DPPH)自由基清除法测定EEBM的抗氧化活性。建立高血脂小鼠模型,将小鼠随机分为正常对照组、模型组、阳性对照组和低、中、高剂量EEBM组,分别采用预防性给药和治疗性给药28和42d。实验结束后,分别测定小鼠血清TC、TG、LDL-C和HDL-C水平。结果:体外抗氧化实验,EEBM对DPPH自由基清除活力的半数抑制率(IC50)为(35.12±0.48)mg·L^(-1),对铁离子还原的半最大效应浓度(EC50)为(148.35±1.38)mg·L^(-1)。降血脂活性,无论是预防性给药还是治疗性给药,与模型组比较,低、中和高剂量EEBM组小鼠血清TC、TG和LDL-C水平均明显降低(P<0.05或P<0.01),HDL-C水平均明显升高(P<0.01)。结论:EEBM可改变高脂血症小鼠血脂质水平使之接近正常水平,EEBM具有明显的降血脂活性。Objective: To discuss the influence of ethanol from Epimedium brevicornu Maxim (EEBM) in the levels of total cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C), and high- density lipoprotein-cholesterol (HDL-C) of the mice with hyperlipemiad, and to clarify its antioxidant activity. Methods: The antioxidant activity of EEBM was evaluated using ferric-reducing antioxidant power assay and DPPH assay, The hyperlipidemic mouse models were established. The mice were randomly divided into normal control group, model group, positive control group, and low, medium, and high doses of EEBM groups prophylactic administration and remedial administration were evaluated in this study. At the end of the experiment, the serum levels of TC, TG, LDL-C, and HDL-C were detected. Results: The IC50 of EEBM to DPPH free radical scaveging activity was (35.12±0.48) mg·L01 in the in vitro antioxidant assay, and its EC50 to ferric-reducing antioxidant power was (148. 35 ± 1.38) mg·L-1. And no matter prophylactic administration or remedial administration, compared with model group, the levels of serum TC, TG, and LDL-C in different doses of EEBM groups were decreased (P〈0.05 or P〈0.01), and the HDL-C levels were increased (P〈0.01). Oonclusion: EEBM can significantly change the plasma lipid levels to near the normal. EEBM has the function of lowering the blood lipid activities in the hyperlipidemia mice.
