奥美拉唑增强奥沙利铂对胃腺癌SGC-7901细胞毒性作用的体外实验

摘　　要：目的观察奥沙利铂(OXA)与囊泡型质子泵(V-ATPase)非特异性抑制剂奥美拉唑(OME)在体外对人胃腺癌细胞株SGC-7901增殖的影响。方法 OME浓度为10、20、40、80、160、320μg/ml分别预处理SGC-7901细胞24 h、48 h后采用MTT法检测奥沙利铂对SGC-7901细胞生长的影响;碘化丙啶(PI)单染流式细胞仪检测胃腺癌SGC-7901的凋亡。结果体外实验表明奥美拉唑联合奥沙利铂可显著抑制SGC-7901细胞的生长,且联合用药效果优于单用奥沙利铂。结论奥美拉唑与奥沙利铂联合使用在体外可显著抑制SGC-7901细胞的生长。Objective To observe whether oxaliplatin combination omeprazole（OME） ,a nonspecific inhibitor of V- ATPase,can enhance the cytotoxicity of gastric cancer cells in vitro experiment. Methods SGC-790l cells were pretreated with omeprazole concentration of 10,20,40,80,160 and 320 μg/ml for 24 h,48 h,The effect of oxaliplatin on the growth of SGC-7901 ceils was detected by MTI＂ assay;detected the apoptosis of gastric cancer SGC-7901 by PI. Results Vitro experi- ment showed that omeprazole combined oxaliplatin could significantly inhibited the growth of SGC-7901 cells, and combination effect was better than oxaliplatin into alonely. Conclusion Omeprazole combined with oxaliplatin has significient effects in in- hibiting the growth of SGC-7901 cells in vitro experiment.
