摘  要:三华李果实富含红色素,对其红色素的主要成分进行了定性和定量测定,明确其结构和含量,为三华李的加工利用提供参考。采用Wonda Sil C18色谱柱(4.6 mm×250 mm,5μm),流动相A:乙腈,B:4%甲酸水溶液,梯度洗脱程序:0 min^25min,6%A^16%A。检测波长520nm,柱温箱40℃,流速1m L/min。试验结果表明三华李果实中红色素主要为矢车菊素-3-葡萄糖苷和矢车菊素-3-芸香糖苷,矢车菊素-3-半乳糖苷未检出。外标法定量,前二者的含量分别为87.26mg/kg和32.41 mg/kg。两种花色苷在0.000 5 mg/m L^0.100 0 mg/m L范围内线性关系良好,标准曲线的决定回归系数R2分别为0.999 9和0.999 8。两种花色苷的加标回收率均大于95%,相对标准偏差(RSD)分别为1.93%和2.40%。样品重复性好,进样峰面积相对标准偏差小于等于1.23%。本方法简便可行,结果可靠,适用于三华李花色苷成分和含量的测定。The Sanhua plum is rich in red pigment. Qualitative and quantitative determination of main composition of the red pigment were carred out.Then their structure and content were known,which can provide refer ence for Sanhua plum processing.The samples were detected with Wonda Sil C18 chromatographic column(4.6 mm×250 mm,5 microns)under 40 ℃ column temerature and 520 nm detection wavelength. The mobile phase consisted of acetonitrile(A)and 4 % formic acid aqueous solution(B)with linear gradient program of 6 %-16 %(A)in 0 min-25 min,at a flow rate of 1 m L/min. Cyanin-3-glucoside and cyanin-3-rutinoside contents of Sanhua plum were 87.26 mg/kg and 32.41 mg/kg,cyanin-3-galacoside was not checked out. The calibration curves of cyanidin-3-glucoside and cyanin-3-rutinoside were linear in the range of 0.000 5 mg/m L-0.100 0 mg/m L,with 0.999 9 and 0.999 8 R^2 respectively. Standard addition recovery rate is more than 95 % and relative standard deviation were 1.93 % and 2.40 % respectively. Sample relative standard deviation of peak area is less than or equal to 1.23 %,which showed good repeatability. This method is simple and reliable. It's suitable to detect anthocyanins component and content in San hua plum.
