摘 要:目的利用常规MRI对阿霉素脂质体治疗大鼠C6胶质瘤的早期疗效进行评价。方法 24只雄性SD大鼠通过脑立体定向仪于右侧尾状核接种l0ul(细胞数为1×106个)C6胶质瘤细胞。在第1周检查均确认有肿瘤生长后分为治疗组(15只)和对照组(9只)。治疗组尾静脉隔天注射10ul/g阿霉素脂质体,对照组尾静脉隔天注射10ul/g生理盐水。分别于接种后第1、2、3周行MRI常规检查,第3周检查后行安乐死进行病理学检查。对照组与治疗组间不同时间肿瘤体积差异的两两比较采用独立样本的秩和检验。结果 24只大鼠均见肿瘤生长,成瘤率为100%。对照组与治疗组大鼠C6胶质瘤的体积第2周时分别为87.92mm3及109.90mm3,2组间肿瘤体积差异无统计学意义(Z=-0.51,P>0.05);第3周时分别为164.85mm3及33.49mm3,2组间肿瘤体积差异有统计学意义(Z=-3.46,P<0.01)。结论阿霉素脂质体治疗大鼠C6胶质瘤是有效的;治疗组较对照组肿瘤内部坏死更加明显,细胞数量明显减少。Objective To evaluate the use of MRI for early detection of tumor response to Liposomal doxorubicin therapy in a rat C6 gfioma model. Methods Twenty-four male SD rats with C6 tumor cells implanted into the stria tum were examined by a 3.0 T MRI scanner, then the rats beating tumors were divided into two groups, treatment group (n= 16) and control group ( n = 9). Rats in the treatment group received liposome doxorubicin therapy through the tail vein every other day, while control group received normal saline instead. Conventional MRI examinations were acquired on 1. 2. 3 weeks after implantation with a 5-inch surface coil. All rats were sacrificed to pe(form the histopathologieil iexamination after MRI examination in the third week. Pretreatment and post treatment tumor.volumes wereicaleulated. Rank sumtest was employed for statistical analysis. Results On MRI, 24 rats demonstrated tumors on day 7 with asuecessful rate of 100%. On week 2. the tumor volumes of the control and treatment group were 87.92 and 109. 90 mm, with no significant difference ( Z = 43.14. P 〉 0.05 ). On week 3. the tumor vol- umes of the controls and treatment group were 156.64 and 29.64 mm3 . and a significant difference was observed ( Z = -3.46, P 〈 0.01 ). Conclusion Liposomal doxorubicin therapy in a rat C6 glioma model is effective. HE stained slices showed more prominent tumor necrosis and death of tumor cells in the treated rats than the controls.
