摘 要:目的:了解感染所引起的新生大鼠感染性脑组织中Sox17和(髓鞘碱性磷酸酶)MBP的表达。方法:将64只2日龄SD大鼠随机分为对照组、实验组。实验组生后第2~6 d持续腹腔注射脂多糖(LPS)每日0.6 mg/kg(0.01 rnl/g)。对照组则按照相同的注射方式及注射剂量,给予生理盐水,建立新生大鼠脑白质损伤模型。注射后按观察时间点不同分为1 d、3 d、7 d、15 d,4个亚组(n=8)。处死大鼠取出脑白质,HE染色观察脑白质区病理变化,Western Blot法测定各组脑组织中Sox17和MBP蛋白的表达,RT-RCR法检测Sox17和MBPmRNA的表达变化。结果:实验组大鼠生长发育缓慢,HE染色显示脑白质区出现组织水肿,部分核固缩、核碎裂。PCR和Western Blotting结果发现Sox17在生后1 d开始表达,7 d达高峰,随后下降,各时间点实验组与对照组相比差异有统计学意义(P<0.05)。MBP在实验组的表达显著下降,各时间点实验组与对照组相比差异有统计学意义(P<0.05)。结论:生后感染可导致新生大鼠脑白质损伤,MBP表达减少,Sox17表达明显升高,提示Sox17剂量的升高对感染后新生大鼠脑组织可能具有保护性修复作用。Objective;To investigate the expression of Soxl7 in brain tissues of newborn rats with infection.Methods:A total of 64 two-day-old Sprague-Dawley(SD) rats were randomly divided into control group and experimental group.At day 2 to 6 after birth,the rats in experimental group were intraperitioneally injected with 0.6mg/kg of lipopolysaccharid,while the rats in control group were injected with saline.Rats were sacrificed and the cerebral white matter was extracted at day1,day3,day7 and day15 after being injected.Haematoxylin-eosin(HE) staining was performed to observe the pathological changes.The expressions of sox17 and MBP in cerebral white matter were determined by Western blotting and RT-PCR analyses.Results:Steady increase in body weight was seen for control group,but not experimental group.The HE staining results showed that part of the nuclear pyknosis and karyorrhexis in cerebral white matter of experimental group's rat.The levels of Soxl7 mRNA and protein in rats of experimental group increased at day 1 d,reached the peak at d 7,and then gradually declined.The low levels of MBP mRNA and protein were found in experimental group's rat.The difference of Sox17 and MBP expression between the experimental and normal group was statistically significant(P 0.05).Conclusion;Postnatal infections may contribute to cerebral white matter injury in immature rats,and reduce the expression of MBP.The expression of Sox17 in brain tissues of infected rats was very higher,which indicated that Sox 17 may play a protective role in infected brain.
