摘  要:目的分析ΔNp63α基因过表达时人宫颈癌细胞SiHa基因表达谱的变化,筛选受ΔNp63α调控的潜在靶基因。方法利用慢病毒转染技术构建稳定过表达ΔNp63α的转染组细胞株SiHa-ΔNp63α和对照组细胞株SiHa-NC。应用基因芯片技术研究两组细胞株基因表达谱变化,并进行生物学信息分析和实时荧光定量PCR验证。结果基因芯片数据共筛选出差异倍数1.5倍以上的基因共1 405个,其中843个基因在SiHa-ΔNp63α细胞中表达上调,562个表达下调。通过生物信息学分析显示这些基因主要参与细胞生长与周期、信号转导、细胞通讯、细胞黏附、细胞转移和侵袭等功能,涉及的信号通路包括抗原加工提呈、细胞因子与受体相互作用、细胞黏附分子、补体系统等。结论研究ΔNp63α调控的潜在靶基因或信号通路,对探索宫颈癌发生发展的机制有重要意义。Objective To analyze the gene expression profiles in response to ΔNp63α overexpression, and screen the potential target genes or signal pathways regulated by ΔNp63α. Methods To generate ΔNp63α overexpressed SiHa cells (SiHa-ΔNp63α) and the control cells (SiHa-NC), recombinant lentivirus transfeetion was performed. Microarray was applied to detect the change of gene expression profiles, and the results were analyzed with bioinfor- marie software. Quantitative real-time PCR was used to validate the expression levels of selected genes. Results Among the 1 405 differentially expressed genes which were statistically significant, 〉 1.5 fold increase or reduce of gene expression, 843 were up-regulated and 562 were down-regulated in SiHa cells with ΔNp63α overexpression. The genes were mostly involved in cell development, cycle regulation, signal transduction, communication, adhesion, metastasis and invasion, etc. The involved signal pathways consisted of antigen processing and presentation, cytokine-cytokine receptor interaction, cell adhesion, complement and coagulation cascades, and so on. Conclusion The research on the potential target genes or mediated signal pathways regulated by ΔNp63α could be helpful to explain the development of cervical cancer.
