摘  要:目的探讨人CCR9基因在乳腺癌细胞中的生物学功能。方法取扁桃体标本并逆转录为c DNA,扩增CCR9基因,连接至pE GFP-N2质粒,脂质体感染人MCF-7乳腺癌细胞。免疫印迹证实CCR9基因在MCF-7乳腺癌细胞表达。CCK-8方法检测MCF-7乳腺癌细胞增殖。结果免疫印迹证实构建pE GFP-N2-CCR9质粒及CCR9基因在MCF-7乳腺癌细胞中高效稳定表达,且CCR9基因能促进MCF-7乳腺癌细胞过度增殖和侵袭(P<0.05或P<0.01)。结论 CCR9基因促进MCF-7乳腺癌细胞增殖和侵袭。Objective To explore the biological function of human CCR9 gene in breast cancer cells. Methods Tonsil samples were taken and reverse-transcripted into cDNA. CCR9 gene was amplified, and was ligated to pEGFP-N2 plasmid. MCF-7 human breast cancer ceils were infected by liposome. Immunoblotting confirmed the expression of CCR9 gene in MCF-7 breast cancer cells. CCK-8 assay was used to detect the proliferation of MCF-7 breast cancer cells. Results Western blot confirmed the construction of pEGFP-N2-CCR9 plasmid and efficient and stable expression of CCR9 gene in MCF-7 breast cancer cells. CCR9 gene could promote the over-proliferatioin and invasion of MCF-7 breast cancer cells(P〈0.05 or P〈0.01). Conclusion CCR9 gene promotes the proliferation and invasion of MCF-7 breast cancer cells.
