摘 要:建立理冲生髓饮中多糖及水溶性蛋白质质量分数测定的方法.测定理冲生髓饮多糖对葡萄糖的换算因子,利用苯酚-硫酸法测定多糖的质量分数;采用考马斯亮蓝G-250染色法测定理冲生髓饮中可溶性蛋白质的质量分数.多糖标准曲线方程为A=12.839 C-0.008 5(r=0.999 3),蛋白质标准曲线方程为A=1.271 C-0.033 1(r=0.999 2),两者分别在0.01~0.1、0.1~1.0 g/L有良好的线性关系.测得理冲生髓饮中多糖质量分数为8.53%,水溶性蛋白质质量分数为0.35%.该两种方法操作简便、灵敏、快速、准确,重复性好,可用于理冲生髓饮中多糖、水溶性蛋白质的质量分数测定.To establish a method for the determination of polysaccharide and water- soluble protein in Lichong Shengsui Drink( LCSSD),the conversion factor of LCSSD polysaccharide to glucose was obtained with refined polysaccharide,and the content of LCSSD polysaccharide was determined by using phenol- sulfuric method. The content of protein was determined by using the dying method with Coomassie brilliant blue G- 250. The polysaccharide standard curve equation was A = 12. 839 C- 0. 008 5( r = 0. 999 3) and protein standard curve equation was A = 1. 271 0 C- 0. 033 1( r = 0. 999 2). Respectively the linear range was 0. 01 ~ 0. 1 g / L and 0. 1 ~ 1. 0 g / L. The content of polysaccharide was 8. 53%,and the content of protein was 0. 35%. The method is simple,fast,accurate and reproducible,and it should be helpful for the determination of polysaccharide and water- soluble proteins in LCSSD.
