摘 要:目的探讨荜茇酰胺对人肺腺癌A549细胞辐射敏感性的影响。方法体外培养人肺腺癌A549细胞,分别加入1、2、4、8、16μmol/L荜茇酰胺作用24、48、72 h,采用CCK-8法检测各组细胞增殖抑制率,筛选出对A549细胞毒性最低的浓度。将A549细胞分为空白对照组、荜茇酰胺组、单纯照射组以及联合处理组,空白对照组不做处理,荜茇酰胺组加入1μmol/L荜茇酰胺,单纯照射组给予8 Gy X线照射,联合处理组先给予1μmol/L荜茇酰胺作用48 h后再给予8 Gy X线照射。采用CCK-8法检测各组细胞增殖抑制率,流式细胞术观察各组细胞周期及凋亡情况。结果不同浓度荜茇酰胺处理A549细胞24、48、72 h后,1μmol/L荜茇酰胺的细胞增殖抑制率均低于其他各组(P均<0.05)。联合处理组48 h时的细胞增殖抑制率高于单纯照射组和荜茇酰胺组(P均<0.05)。联合处理组G0/G1期细胞比例较单纯照射组增多,而S期细胞比例减少,细胞凋亡率高于单纯照射组(P均<0.05)。结论 1μmol/L荜茇酰胺能够增加A549细胞对X射线的辐射敏感性,其机制可能与促进细胞增殖抑制、诱导细胞凋亡、引起细胞G_0/G_1期阻滞有关。Objective To investigate the effect of piplartine on radio-sensitization of human pulmonary adenocarcinoma A549 cells.Methods A549 cells cultured in vitro were treated with 1,2,4,8 and 16 μmol/L piplartine for 24,48 and 72 h.CCK8 assay was used to measure the proliferation inhibition of piplartine on A549 cells in each group and then we selected the minimum concentration which had no obvious toxic effect on A549 cells.A549 cells were divided into four groups:the control,piplartine group,radiation group and the combination treatment group.The control group was not treated.The piplartine group was given the concentration of 1 μmol/L piplartine,the radiation group was given X-ray radiation of 8 Gy alone,and the combination treatment group was given the concentration of 1 μmol/L piplartine for 48 h,then was exposed to X-ray radiation of 8 Gy.CCK8 assay was used to measure the proliferation inhibition of piplartine on A549 cells in each group.Cell cycle and apoptosis of A549 cells were analyzed by flow cytometry(FCM).Results After being treated with piplartine for 24,48 and 72 h,the proliferation inhibition rate of the piplartine group was lower than that of the other groups(all P〈0.05).The proliferation inhibition in the combination treatment group was higher than that of the radiation group and the piplartine group at 48 h(all P〈0.05).The percentage of cells in the G0/G1 phase of the combination treatment group was significantly increased,and was decreased in the S phase,and apoptosis rate was also higher than that of the radiation group(P〈0.05).Conclusion Piplartine at the concentration of 1 μmol/L can enhance the radiosensitivity of A549 cells,which may be associated with the enhancement of radiation-induced inhibition of cell proliferation and induction of apoptosis as well as cell arrest in G0/G1 phase.
