摘 要:目的研究缬草波春对非炎症性结肠致敏大鼠模型大脑海马旁回(hippocampal convolution)γ-氨基丁酸-B受体(γ-aminobutyric acid-B receptor,GABA-B receptor)表达的影响。方法用正丁酸盐灌肠法诱导非炎症性结肠致敏大鼠模型,设药物实验组、模型对照组及空白对照组3组(n=8),药物实验组用缬草波春对模型组灌胃2周,对照组灌注等量生理盐水,2周后分别处死空白对照组、模型对照组及药物实验组大鼠,取相关海马旁回,用免疫组化半定量的方法检测组织GABA-B受体表达。结果空白对照组(0.235±0.014)、模型对照组(0.161±0.017)及药物实验组(0.238±0.019)的大脑海马旁回有GABA-B受体表达;模型对照组GABA-B受体表达较空白对照组减少,差异有统计学意义(t=2.86,P<0.01);缬草波春使模型GABA-B受体表达上调,与模型对照组比较差异有统计学意义(t=3.28,P<0.01),与空白对照比较差异无统计学意义(t=1.45,P>0.05)。结论非炎症性结肠致敏大鼠模型大脑海马旁回GABA-B受体表达减少,缬草波春使非炎症性结肠致敏大鼠模型大脑海马旁回GABA-B受体表达增加。Objective To study the effects of Valepotriate on the expression of γ-aminobutyric acid- B receptor( GABA-B receptor) in brain parahippocampal gyms in the rat model of noninflammatory colonic hypersensitivity. Methods Rat models of noninflammatory colonic hypersensitivity were induced by receiving enemas of butyrate solution. The rats were divided into experimental group, model control group and bland control group (n = 8). The rats in experimental group received enemas of Valepotriate for 2 weeks, and the rats in control group received enemas of equivalent normal saline. The rats were killed after 2 weeks and the brain parahippocampal gyms was collected. The expressions of GABA-B receptor in the tissues were tested by semiquantitative immunohistochemistry methods. Results The expressions of GABA-B receptor in blank control group,model control group and experimental group were 0. 235 ±0. 014,0. 161 ±0. 017 and 0. 238 ± 0. 019. The expression of GABA-B receptor in model control group was significantly lower than that in blank control group, and the difference was statistically significant( t = 2. 86,P 〈 0. 01) . The expression of GABA-B receptor in experimental group was up-regulated, with the significant difference comparing with the model control group( t = 3. 28, P 〈 0. 01 ) , and without the significant difference comparing with the blank control group (t = 1. 45 , P 〉 0. 05 ) . Conclusion The expression of GABA-B receptor in brain parahippocampal gyms in rat model of noninflammatory colonic hypersensitivity is decreased, and Valepotriates can increase the expression of GABA-B receptor.
