摘 要:目的探讨野生猕猴桃根黄酮提取物对胃癌MKN-49P细胞增殖作用的影响及机制。方法采用CCK-8比色法、流式细胞术、Western Blot法检测野生猕猴桃根黄酮提取物对胃癌MKN-49P细胞增殖作用的影响。结果野生猕猴桃根黄酮提取物对胃癌MKN-49P细胞增殖有明显的抑制作用,且呈时间-浓度依赖性;其浓度升高,细胞凋亡增加,G1期细胞增加,S、G2/M期细胞减少,抑制周期蛋白CyclinD1表达降低。结论野生猕猴桃根黄酮提取物可抑制胃癌MKN-49P细胞增殖、促进细胞凋亡、阻滞细胞周期于G1期,其主要机制可能与抑制周期蛋白CyclinD1表达降低有关。Objective To investigate the effect and mechanism of wild Chinese actinidia root flavonoids extracts in gastric cancer MKN-49 P cell proliferation. Methods Using the CCK-8 assay,flow cytometric( FCM) analysis and Western Blot method to detect the effect of flavonoids on gastric cancer MKN-49 P cell proliferation. Results Wild Chinese actinidia root flavonoids extracts had obvious inhibitory on gastric cancer MKN-49 P cell proliferation in a time-concentration dependence; as concentration increased,apoptosis increased,G1 staging cells increased,S and G2 / M staging cells reduced,protein expression of Cyclin D1 which inhibiting cell cycle reduced. Conclusion Wild Chinese actinidia root flavonoids extracts can inhibit gastric cancer MKN-49 P cell proliferation,promote cells apoptosis,inhibit cell cycle in G1 staging,the main mechanism is probably related with protein expression of Gyclin D1 which inhibiting cell cycle reduced.
