摘  要:目的 :研究富血小板血浆(platelet rich plasma,PRP)对负载人胚胎来源的间充质干细胞(h ESC-MSCs)多孔磷酸钙骨水泥(calcium phosphate cements,CPC)复合物的成骨促进作用。方法:将h ESC-MSCs接种于CPC表面,扫描电镜(SEM)观察h ESC-MSCs在CPC表面的黏附情况。建立直径为8 mm的大鼠颅骨圆形缺损模型,将18只大鼠随机分为3组,分别是CPC+h ESC-MSCs组、CPC+h ESC-MSCs+30%PRP组和CPC+h ESC-MSCs+50%PRP组。术后12周显微CT和H-E染色观察新骨和血管生成情况。应用SPSS16.0软件包对数据进行单因素方差分析。结果:SEM显示,细胞可良好地黏附在CPC支架的表面。显微CT示复合PRP后,CPC材料的降解程度逐渐明显,且新生骨量逐渐增加。H-E染色显示,复合PRP组的新骨生成率和血管密度高于未复合PRP组,但是复合不同浓度的PRP组间无显著差异。结论:浓度为30%和50%的PRP与负载h ESC-MSCs的CPC复合后,均可促进骨缺损区新骨的生成。PURPOSE: To investigate the effects of macroporous calcium phosphate cement(CPC), human embryonic stem cells(h ESC) and platelet-rich plasma constructs for bone regeneration in vivo in critical-sized cranial defects in rats.METHODS: Three groups were tested in rats:(a) CPC with h ESC-derived mesenchymal stem cells(CPC+h ESC-MSCs);(b) CPC with h ESC-MSCs and 30% platelet rich plasma(PRP)(CPC+h ESC-MSCs+30%PRP) and(c) CPC+h ESC-MSCs+50%PRP. 18 rats were sacrificed 12 weeks after operation. Micro-CT and bone histomorphologic analysis were conducted to compare the status of new bone formation. SPSS16.0 software package was used for one way analysis of variance.RESULTS: At 12 weeks in vivo, the groups with h ESC-MSCs and PRP had more new bone formation compared with other two groups; however, there was no significant difference in new bone area fraction between different concentration of PRP groups. CONCLUSION: Macroporous CPC with h ESC-MSCs and PRP may be useful for bone regeneration in craniofacial and orthopedic fields.
