摘  要:目的:研究感觉神经对大鼠下颌切牙釉质和牙本质形成的调控作用。方法:将48只成年SD大鼠随机均分为失右侧感觉神经(截断下牙槽神经)组(IANx组)、失右侧交感神经(摘除颈上神经节)组(SCGx组)、失右侧感觉神经和交感神经组(IANx+SCGx组)、假手术组(sham组)(n=12),术后2周HE染色法观察各组大鼠的下颌切牙结构。将10只4周龄SD大鼠随机均分为IANx组和sham组(n=5),术后2周体外分离培养其牙髓干细胞(r DPSCs);经流式细胞术鉴定后,比较其克隆形成和增殖能力,并用RT-PCR检测釉质和牙本质发育相关基因Amg、Enam、Dspp、Dmp1 mRNA的表达水平。结果:IANx组和IANx+SCGx组大鼠的右侧下颌切牙发生明显的白垩色改变,釉质和牙本质的厚度均明显低于sham组(P<0.05)。IANx组大鼠的r DPSCs与sham组相比,克隆形成和增殖能力均明显下降(P<0.05),釉质和牙本质发育相关基因的表达水平也明显降低(P<0.05)。结论:感觉神经可通过影响r DPSCs的生物学行为而影响釉质和牙本质的形成。AIM: To investigate the role of sensory nerve in the formation of enamel and dentin in incisor of rats. METHODS : 48 adult SD rats were divided into 4 groups (n = 12) randomly. The rats received unilateral sensory nerve sec-tion (IANx) , unilateral sympathetic nerve section (SCGx) , unilateral sensory nerve and sympathetic nerve section (IANx + SCGx) , and sham operation, respectively. The rats were sacrificed at the end of 2 W after operation and HE staining was performed to observe the changes of incisors. 10 SD rats (4 W of age) were randomly divided into IANx group and sham group (n =5). rDPSCs were cultured and identified from the 2 groups of rats 2 W afte operation. The colony - forming abili-ty, proliferation rate and expression level of genes associated with enamel and dentin formation were compared between the 2 groups. RESULTS : The incisors of IANx and IANx + SCGx groups turned chalky 2 W after denervation. HE staining showed reduced enamel and dentin formation (P 〈 0.05). The rDPSCs of IANx group showed lower clonogenic and prolifera-tive potential (P 〈 0.05) and expressed lower levels of Amelogenin, Enamelin, Dspp and Dmpl mRNA than sham group (P 〈0.05). CONCLUSION: Sensory nerve supports the formation of enamel and dentin by regulating rDPSCs.
