摘 要:DNA转座子可以持久、有效地改变哺乳动物以及其他生物物种的基因组,是一类可以改变在其自身基因组中所处位置的遗传因子。piggy Bac转座系统是一种目前常用的哺乳动物基因工程研究工具,用于癌基因发现、转基因动物制备、体内基因转移以及体外细胞的基因修饰如多能性干细胞诱导分化等。此外,piggy Bac转座子还具有宿主基因组无痕剪切、目的DNA序列靶向整合等多种特性,因而在构建动物模型和基因治疗中得到了广泛应用。作为一种新的分子遗传工具,piggy Bac转座系统将在医学和生命科学研究中发挥越来越重要的作用。DNA transposon is a kind of factor that is able to translocate gene in its genome, thus offering an efficient method for permanently modifying the genome of mammals. The piggyBac (PB) transposon system has been proven effective in mammalian genomic engineering, including cancer gene discovery, animal transgenesis, in vivo gene delivery as well as in vitro genetic modificatioii like induced pluripotent stem cells. In additionj has many desirable features, such as seamless excision of transposons from the genomic DNA and the potential to target integrationevents to desired DNA sequences. Therefore, the piggyBac translocation system is an ideal genetic tool in the construction o f animal models and gene therapy, and we can anticipate that the piggyBac transposon system will play a more and more important role in biomedical research.
