摘 要:目的探讨卡芬太尼合成方法的研究进展,以期为该类药物的进一步应用和新型高效药物的研发提供借鉴。方法以芬太尼类衍生物药物研发为线索,查阅了1976年至今的国内外文献,对卡芬太尼的合成进展进行综述。结果卡芬太尼的合成主要有3条路线,分别为以1-苄基-4-哌啶酮、1-苯乙基-4-哌啶酮和N-Boc保护哌啶酮为原料的合成方法。结论卡芬太尼工艺路线的比较和分析为卡芬太尼衍生物的合成、生产以及工艺选择提供了依据。To review the research development of synthetic method of carfentanilin in order to develop a new efficient drugand provide further application of this kind of drugs. Focused on the drug development of fentanyl derivatives, the litera-tures about the synthetic method of carfentanil were reviewed, which were reported by various research groups since 1976.The preparation of crafentanil included 3 synthetic methods, by using 1-benzyl-4-piperidone, 1-phenethyl-4-pipafidone and N-boc-protected piperidone as starting materials^respectively. The comparison and analysis of the above synthetic methods canprovide the suggestion on the preparation of carfentanil and its derivatiyes, as well as the optimization of production process.
