摘  要:目的采用D-最优设计响应面法,优选脑得生方提取工艺。方法以脑得生方提取工艺的关键参数乙醇浓度、乙醇用量和提取时间为自变量,进行D-最优设计;采用可见分光光度法和高效液相色谱法,测定脑得生方中主要指标成分总皂苷、总黄酮和阿魏酸的含量,并以它们的加权含量为因变量,进行响应面分析,优选出脑得生方提取的最佳工艺。结果以10.81倍用量、浓度为80%的乙醇回流提取45 min,作为提取的最佳工艺。结论利用D-最优设计响应面法能够优选脑得生方提取工艺参数,提高了有效成分的提取效率,有助于脑得生方的质量标准研究。Objective D-optimal response surface methodology is used to optimize the extraction process of Naodesheng prescription. Methods With the key factors of extraction process as the independent variables,including ethanol concentration,volume and extracting time,we conducted the D-optimal design. Based on the contents of total ginsenosides,total flavonoids and ferulic acid determined by visible spectrophotometric method and high performance liquid chromatography,their weighted content was calculated and adopted as the dependent variable for the response surface methodology to optimize the extraction process of Naodesheng prescription. Results The optimal factors were 10. 81 times volume of 80% ethanol and 45 minutes of reflux extracting time. Conclusion D-optimal response surface methodology can be used to optimize the extraction process of Naodesheng prescription,which helps improve the extraction efficiency of active components,and contribute to the quality standard research of Naodesheng prescription.
