摘  要:目的:研究石榴的干燥种子等11种蒙药在体外对马拉色菌的抑制活性,并研究两种蒙药合用对马拉色菌临床株和标准株的抑制活性的变化。方法:用琼脂稀释法、琼脂柱法筛选出最佳抗糠秕马拉色菌的一种蒙药,然后用该种蒙药与其它蒙药按等比例混合后对5种马拉色菌的标准株和临床株进行药敏试验。分析筛选出的蒙药石榴种子在体外对糠秕马拉色菌存活率、脂肪酶活性、脂质利用活性、诱导人角质细胞系分泌部分细胞因子活性的影响、并在兔模型上观察了石榴种子治疗马拉色菌毛囊炎的临床疗效。结果:石榴种子的醇提液抑制糠秕马拉色菌的作用最强,其MIC为2.5mg/ml。石榴种子与其他蒙药联合对马拉色菌的抑制作用显著增强,MIC最低达到0.313mg/ml。石榴种子在体外能够抑制糠秕马拉色菌的存活率,并降低其脂肪酶活性,抑制其脂质代谢活性。石榴种子抑制了糠秕马拉色菌诱导人角质细胞系分泌IL-1β、IL-6和TNF-α的活性。石榴种子外用治疗马拉色菌毛囊炎具有明显疗效。结论:蒙药醇提液中以石榴种子的抑菌效果最佳,石榴种子对马拉色菌的抑制作用具有临床应用的前景。Objective: To evaluate the antifungal efficacy of Pomegranate and other traditional Mongolian medicine. Methods: Through the way of agar dilution and agar column, the efficacy of gradient concentrations of solutions inhibiting Malassezia furfur were observed. The influence of Pomegranate was further analyzed on cell viability, lipase activity and lipid utilization activity of Malassezia furfur in vitro. Together, the inhibitory effect of Pomegranate on Malassezia-induced cytokines secretion by HaCaT was evaluated. Finally, the topical effect of Pomegranate on Malassezia folliculitis was observed in rabbit model. Results: The active ingredients extracted by alcohol were stronger than those extracted by distilled water in terms of antifungal efficiency and spectrum. The inhibitory effect of alcohol extract from Pomegranate on Malassezia furfur is the strongest among all the Mongolian medicines, and the MIC is 2.5mg/ml. Besides, the inhibitory effect of Pomegranate combined with other Mongolian medicines on Malassezia spp increased significantly, and the MIC is as low as 0. 313mg/ml. The cell viability, lipase activity and lipid utilization activity of Malassezia furfur were significantly inhibited by Pomegranate in vitro. Besides, Malassezia-induced IL-1β, IL-6 and TNF-α secreted by HaCaT was significantly suppressed by Pomegranate. Satisfactory effect was achieved by Pomegranate on Malas-sezia folliculitis. Conclusion: Among the 11 traditional Chinese Mongolian medicines, Pomegranate had stronger inhibitory effect against Malassezia spp. The inhibitory effect of Pomegranate on Malassezia spp makes it a promising clinical application in future.
