摘 要:乙型肝炎病毒(hepatitis B virus,HBV)是一种广泛存在的人类病原体,可引起肝炎、肝硬化和肝细胞癌(hepatocellular carcinoma,HCC)。HBV感染严重威胁人类的健康,尤其以我国和东南亚国家为甚。慢性乙型肝炎和因慢性HBV感染而导致的HCC的发病机制、诊断和治疗仍是一个很具挑战性的课题。目前研究病毒性肝炎以及肝炎后肝硬化和HCC的发病机制和诊治很大程度上依赖于动物模型,而良好的动物模型的选择是决定研究结论是否可靠的基础。土拨鼠是一种主要生活在北美大平原的动物,其个体和肝脏大小特征、携带的土拨鼠肝炎病毒(woodchuck hepatitis virus,WHV)的致病机制,疾病进展与人HBV相似性等因素很利于研究乙肝病毒性肝炎和HCC的发生和发病机制、诊断与治疗。因此,通过土拨鼠肝炎动物模型能更好地了解HBV与HCC的复杂关系并将促进更好的预防和治疗新策略的制定。本文就土拨鼠动物模型在乙型病毒性肝炎以及HCC研究中的应用进展做一概述。Hepatitis B virus(HBV) is a widespread human pathogen that can cause hepatitis,cirrhosis, and hepatocellular carcinoma(HCC). HBV infection has been a serious threat to human health,especially in China and Southeast Asian countries. The pathogenesis, diagnosis and treatment of chronic hepatitis B and HBV caused HCC are still very challenging. Currently, understanding of the pathogenesis and diagnosis of viral hepatitis, liver cirrhosis post hepatitis, and HCC are largely dependent on the animal model system, and the choice of a well-established animal model is the basis in accomplishment of the study. The features of woodchuck body weight, the size of its liver, the pathogenesis of the hepatitis B virus it carries, and the progress of the disease course are very similar to those of human hepatitis B virus, thus, the woodchuck is a very useful animal model in hepatitis B virus infection and in the study of the development, pathogenesis,diagnosis, and treatment of hepatitis B and HCC. Therefore, the woodchuck hepatitis B virus animal model enables us to have a better understanding of the complex relationship between HBV infection and HCC. It will also promote a better prevention and treatment strategy for hepatitis B infection. In this paper, recent reports in the application of the woodchuck animal model in the field of hepatitis B and HCC will be summarized.
