摘 要:[目的]探讨FMNL2基因沉默对人大肠癌细胞体内外增殖及侵袭能力的影响。[方法]筛选并建立FMNL2基因沉默的SW80/M5细胞系;荧光定量RT-PCR和Western blot检测干扰效率;CCK8法检测FMNL2对SW80/M5增殖能力的影响;流式细胞仪检测细胞周期;侵袭小室实验检测其对SW80/M5侵袭能力的影响;裸鼠皮下成瘤实验和脾注射法体内转移实验检测FMNL2基因表达沉默对SW80/M5在体内增殖、侵袭能力的影响。[结果]FMNL2基因沉默后显著抑制SW80/M5在体内外增殖、转移及侵袭能力(P<0.05)。[结论]FMNL2与大肠癌多种恶性生物学功能的调控密切相关,可能是潜在的治疗靶点。[Objective] To investigate the effect of FMNL2 gene silence on the proliferation of invasion of human colorectal cancer(CRC) cells in vivo and in vitro. [Methods] Human colorectal cancer SW80/M5 cells were transfected with FMNL2 specific sh RNA plasmid vectors,and FMNL2- silenced cells were screened by G418. Quantitative RT-PCR and Western blot analysis were used to detect the expressions of FMNL2 m RNA and protein,respectively. The cell proliferation was determined by CCK8 assay,the cell invasion ability was detected by boyden invasive assay in vitro. Tumorigenesis and metastasis in vivo were examined by whole-body visualizing hepatic metastasis animal model in nude mice. [Results] FMNL2 silencing inhibited cell proliferation,migration and invasion(P〈0.05) in vitro and in vivo.[Conclusion] FMNL2 gene may have multiple biological functions during colorectal cancer progression and it may used as a novel therapeutic target against CRC.
