摘　　要：[目的]观察PIAS3在结肠癌组织及细胞中的表达,探讨PIAS3在结肠癌发生发展中的作用。[方法]采用免疫组织化学染色法检测60例结肠癌及同源癌旁组织中PIAS3蛋白的表达,实时荧光定量PCR(q RT-PCR)、Western blot检测人结肠癌SW480、SW620及COLO205细胞株中PIAS3 m RNA及蛋白表达水平。[结果 ]结肠癌组织中PIAS3蛋白表达显著高于癌旁正常组织(χ~2=8.543,P=0.006);且表达强度随肿瘤病理分化程度的降低、Dukes分期的增高及肿瘤体积的增大而逐渐增高,差异有统计学意义(P均<0.05);PIAS3在结肠癌SW480、SW620及COLO205细胞株中均有表达,其中COLO205细胞株中PIAS3蛋白及m RNA表达高于SW480和SW620细胞株(P<0.05)。[结论 ]PIAS3的表达水平以及活性与结肠癌的发生发展有关,可能为结肠癌的早期诊断及靶向治疗提供新的分子靶点。[Objective] To investigate the expression of PIAS3 in colon cancer and its significance.[Methods] The expression of PIAS3 protein was detected by immunohistochemisry（IHC） in60 samples of colon cancer tissue and corresponding adjacent normal colon tissue. PIAS3 protein and m RNA were detected in human cancer cell lines SW480,SW620 and COLO205 by q RT-PCR and Western blot,respectively. [Results] The expression level of PIAS3 protein was significantly higher in colon cancer tissue than that in adjacent non-cancer tissues（χ~2=8.543,P=0.006）. The expression of PIAS3 was closely correlation with differentiation,Dukes stage and size of colon cancer（P 〈0.05）. The positive expression of PIAS3 was observed in SW480,SW620 and COLO205 cells,the level of expression in COLO205 cells was higher than that in others（P〈0.05）.[Conclusion] The expression of PIAS3 may be related to the occurrence and development of colonic neoplasm,and it might be a new biomarker or target for diagnosis and treatment of colon cancer.
