microRNA-371-5p在结肠癌中的表达及其临床意义

摘　　要：目的通过检测microRNA-371-5p在结肠癌患者血浆中的表达变化并探讨其临床意义。方法选取结肠癌患者血浆50例作为实验组,另选择年龄、性别及种族与实验组相匹配的健康体检者的血浆30例作为健康对照组,采用实时荧光定量聚合酶链反应(qRT-PCR)检测血浆中microRNA-371-5p的表达水平并分析其在不同临床病理资料间的差异。应用受试者工作特征曲线(ROC)分析并计算曲线下面积(AUC),并利用曲线下面积(AUC)估计肿瘤标志物的诊断价值。结果 microRNA-371-5p在结肠癌患者血浆中的相对表达量为3.03±0.55,显著低于健康体检者血浆中的相对表达量(3.98±0.47),差异具有统计学意义(t=-4.93,P<0.05);结肠癌患者血浆中的microRNA-371-5p与年龄、性别以及CEA水平无相关性,而与临床病理分级(TNM分级)、淋巴结转移与否成正相关(P<0.05)。miR-371a-5p作为结肠癌生物标志物(AUC)的曲线下面积为0.894,特异度为87%,灵敏度为86%。结论 microRNA-371-5p在结肠癌患者血浆中呈低表达水平,推测microRNA-371-5p可能参与结肠癌的发生发展过程,有望成为预测结肠癌的检测指标之一。Objective To investigate the expression difference and clinical significances of microRNA-371-5p in serum of patients with colorectal cancer. Methods The expression levels of microRNA-371-5p in the plasma of 50 colorectal canc- er patients and 40 healthy controls were detected by qRT-PCR, and its correlation with clinical pathological parameters was analyzed. Receiver operating characteristic curve （ROC） analysis and the area under curve （AUC） was performed to estimate the diagnostic value of tumor markers. Results The expression level of microRNA-371-5p in coloreetal cancer plasma （3.03 ± 0.55 ） was significantly lower than that in the in serum of healthy people （3.98 ± 0.47）, t = - 4.93, P 〈 0.05. The expression level of microRNA-371-5p in colorectal cancer plasma was well correlated with the TNM stages and the metastasis of lymph node （ P 〈 0.05 ）, but not with the age, gender and CEA level （ P 〉 0.05 ）. AUC was 0. 894, the specificity was 87% ,and the sensitivity was 86%. Conclusion The microRNA-371-5p presents a low expression in the patients with colorectal cancer, so it might be involved in initiation and progression of colorectal cancer, thus it might be one of biomarkers of colorectal cancer.
