摘  要:目的探讨丁苯酞(3-n-Butylphthalide,NBP)对Aβ1-42致阿尔茨海默病模型大鼠记忆的影响作用与可能机理。方法将Aβ1-42制备的AD大鼠随机分为五组,对照组,AD模型组、NBP 40mg/kg组、NBP 80mg/kg组和NBP 160mg/kg组,20只/组,对照与模型组给予蒸馏水,给药30d。结果对照组大鼠状态良好,AD模型组大鼠精神萎靡,活动迟缓,进食量少,体质量增高不明显,NBP组大鼠的状态介于模型和对照组间。与对照组比,AD模型组逃避潜伏期延长,SOD和OH-降低,胆碱酯酶增高(P<0.05)。NBP组可明显降低AD大鼠逃避潜伏期,增高SOD和OH-,降低胆碱酯酶(P<0.05),与NBP低剂量组比,NBP中、高剂量可明显降低逃避潜伏期时间、增高OH-、降低胆碱酯酶(P<0.05)。与对照组比,AD模型组的Tau表达明显增高(P<0.05),AKT表达明显降低(P<0.05),与AD模型组比,NBP组的Tau表达明显降低(P<0.05),AKT表达明显增高(P<0.05)。结论 NBP可明显改善AD大鼠的记忆能力,可增高脑内SOD和OH-,降低胆碱酯酶,改善脑内Tau和AKT表达。Objective To investigate the effect of 3-n-Butylphthalide (NBP) on memory ability of Aβ1-42 induced Alzheimer's disease (AD) rat's memory and the possible mechanism. Methods AD rats were randomly divided into 4 groups., the AD model group, NBP 40 mg/kg group, NBP 80 mg/kg group, and NBP160 mg/kg group. Results In control group, rats were in good condition, rats in AD model group showed acting slowly, eating less. The condi tions of rats in 3 NBP groups were between the AD model and the control group. Compared with the control, escape latency had increased, SOD and OH- had decreased, cholinesterase had increased (P〈0.05). 3 NBP groups can significantly reduced AD escape latency, increased SOD and OH-, decreased cholinesterase enzyme (P〈 0.05). Compared with NBP40 mg/kg group, NBP80 mg/kg and 160 mg/kg groups can be significantly reduced escape la- tency time, increased OH- , decreased cholinesterase (P〈0.05). Compared with control group, Tau in AD model group had significantly increased (P〈0.05), AKT and BNDF had significantly decreased (P〈0.05). Tau in 3 NBP groups had significantly decreased (P〈0.05), AKT and BNDF had significantly increased (P〈0.05). Con- clusion NBP can significantly improve ADrs memory. In which, NBP can increase SOD OH- , AKT and BNDF ex- pression, decrease cholinesterase and Tau expression.
