红藻氨酸致癫痫大鼠脑内炎症反应的研究

摘  要:目的通过观察大鼠痫性发作后血清和脑脊液中炎症因子白细胞介素-6(Interleukin-6,IL-6)水平的动态变化,进而探讨炎症反应与癫痫的关系。方法侧脑室注射红藻氨酸(Kainic acid,KA)建立颞叶癫痫大鼠动物模型,用ELISA方法测定大鼠痫性发作后6小时、24小时、72小时、1周血清和脑脊液中IL-6的水平,并与对照组相比较。同时观察癫痫发作后神经元的病理变化。结果血清和脑脊液中IL-6水平在癫痫发作后6小时逐渐增加,24小时达高峰,以后逐渐下降。致痫24小时后出现大量神经元坏死。结论炎症反应参与了癫痫后神经元损伤。IL-6或许可作为癫痫脑损伤后的早期外周生化标志物。Objectives To investigate the levels of IL- 6 in serum and cerebrospinal fluid(CSF) of rats with epilepsy,and then to approach the relationship between inflammatory and epilepsy. Methods An animal model of epilepsy was established in Male SD rat by intracerebroventricular injection of KA. The levels of IL- 6 in serum and CSF was measured by ELISA at 4 different time points—6,24,72 hours and 1 week after seizures. And control group were set up to be contrasted and be observed. At the same time,the pathological changes of neuron were observed after seizures. Results In the experimental group,levels of IL- 6 protein in both serum and CSF increased gradually 6 hours after seizures and declined gradually after reaching their highest level at the 24 hour time point. A large amount of of neuron necrosis in hippocampus appeared at 24 hours after seizures. Conclusion Inflammatory reaction maybe involve in seizure- induced brain damage. IL- 6 may be a key biomarker for predicting early seizure- induced neuron damage and necrosis.
