摘 要:目的通过观察Apo-E(-/-)小鼠主动脉根部血管MMP-9的表达情况,进一步了解MMP-9与动脉粥样硬化斑块形成过程的相关性。方法本实验采用免疫组织化学和逆转录聚合酶链技术对MMP-9在基因敲出小鼠动脉中的表达进行研究。结果正常小鼠冠状动脉中MMP-9有微量表达,动脉粥样硬化斑块中检测到MMP-9 mRNA和MMP-9蛋白的表达明显增高。结论 MMP-9可能参与小鼠动脉粥样硬化病变过程,并在小鼠主动脉根部血管壁斑块中表达增高。Objective To investigate the expression of matrix metalloproteinase-9 (MMP-9)in the aor- tic root of Apo-E(-/-) mice and to explore the correlation between MMP-9 and atherosclerotic plaque forma- tion. Methods The expression of MMP-9 in the arteries of knockout mice was studied by immunohistochernis- try and reverse transcription polymerase chain reaction (RT-PCR). Results The expression of MMP-9 and MMP-9 was significantly increased in the coronary arteries of normal mice. The expression of MMP-9 mRNA and MMP-9 protein was significantly increased in atherosclerotic plaques. Conclusion MMP-9 may be in- volved in the process of atherosclerosis in mice, and the expression of MMP-9 in vascular wall plaque of aortic root in mice is increased.
