Establishment and Evaluation of Spontaneous Recurrent Seizures in Rat Amygdala Rapid Kindling Epilepsy Model

摘  要:目的:建立易反复自发(SRS)的大鼠杏仁核快速电点燃癫痫(EP)模型。方法:40只SD大鼠随机地分为对照组(n=10)、SRS EP模型A组(n=15,模型A组)及SRS EP模型B组(n=15,模型B组);对照组大鼠只植入电极,不予刺激;模型A组大鼠给予60 Hz频率、1.0 ms波宽、0.5 m A强度及10 s串长刺激1 min;模型B组大鼠给予16 Hz频率、1.0 ms波宽、0.8 m A强度及10 s串长刺激20 min;两组均10次/d,连续5 d;于刺激1月后给予同样的刺激强度进行刺激后,观察3组大鼠的脑电图(EEG)变化,EP发作和SRS时EEG的频率和波幅、EP点燃过程中大鼠的行为学表现、EP点燃大鼠EP发作的稳定性和持久性及SRS发作率。结果:模型A组和B组大鼠点燃EP的成功率分别为80%和87%,差异无统计学意义(P>0.05),模型B组SRS发作率(69.2%)高于A组(25%,P<0.05);对照组大鼠EEG频率和波幅分别低于两模型组EP发作时的频率和波幅(P<0.05);两模型组大鼠在EP发作时和SRS时的EEG频率、波幅比较,差异无统计学意义(P>0.05);随着刺激次数增加及时间的推移,两模型组大鼠先出现洗脸、闭眼及点头运动等表现,后逐渐出现前肢抽动、痉挛,最后直至出现站立、跌倒的Ⅳ~Ⅴ级的大发作。结论:采用16 Hz频率、1.0 ms波宽、0.8 m A强度及10 s串长刺激大鼠杏仁核20 min点燃的EP模型更容易出现SRS发作。Objective:To establish a spontaneous recurrent seizure in rat amygdaloid kindling epilepsy model.Methods:40 SD rats were divided into three groups:control group(n = 10),Model A of SRS EP(n = 15,Group A) and SRS EP Model Group B(n = 15,Group B) :rats in the control group only implant electrode without stimulating; Group A rats were stimulated by frequency of 60 Hz FWHM of 1.0 ms,intensity of 0.5 m A,duration of 10 s strings long stimulation for 1 min; Group B rats were stimulated by frequency of 16 Hz,FWHM of 1.0 ms,intensity of 0.8 m A,duration of 10 s strings long stimulation for 20 min; both groups were stimulated 10 times/d for consecutive 5 d.One month after stimulation,stimulating the rats with same intensity of stimulation,observing frequency and amplitude of EEG changes while SRS and EP,observing behavioral manifestations of rats while EP kindling process,observing stability and duration and SRS attack rate while rats EP attacked while kindling.Results:The rate of being kindled from group A and group B was 80% and 87%,the differences showed no statistical significance(P〉0.05).The rate of spontaneous recurrent seizures of group B(69.2%) was higher than group A(25%),differences showed statistical significance(P〈0.05).EEG frequency and amplitude of control group rats were lower than that of EP attacked model groups(P〈0.05); when the rats have epileptic seizure,comparing with the frequency and amplitude of SRS,differences showed no statistical significance(P〈0.05); with the increase of stimulation times and processing of time,both Model groups rats were observed with movements such as face washing,eye closing and nodding head,then gradually forelimbs twitching,cramp and in the end they were observed with Ⅳ to Ⅴ level of generalized tonic-clonic seizures like standing up and falling down.Conclusion:Adopting frequency of 16 HZ,FWHM of 1.0 ms,intensity of 0.8 m A,duration of 10 s strings long stimulation for 20 min on rats' amygdala kindled EP model co
